Description
Overview of the medicine
Abazino 150 mg Tablet contains Abazin, an Angiotensin II Receptor Blocker (ARB) used to treat high blood pressure (hypertension) and heart failure. It helps to relax blood vessels, lowering blood pressure and making it easier for your heart to pump blood.
Uses & Indications
Dosage
Adults
Hypertension: Initial dose 150 mg once daily, may be increased to 300 mg daily if needed. Heart Failure: Initial dose 75 mg once daily, titrated up to 150 mg or 300 mg daily as tolerated.
Elderly
No initial dosage adjustment is typically required, but monitoring of renal function is advised.
Renal_impairment
For severe renal impairment (CrCl <30 mL/min), initiate with a lower dose (e.g., 75 mg once daily) and monitor closely. Not recommended in patients undergoing dialysis.
How to Take
Abazino tablets should be swallowed whole with water, with or without food, once daily at approximately the same time each day.
Mechanism of Action
Abazin selectively blocks the binding of angiotensin II to the AT1 receptor, which is found in many tissues, including vascular smooth muscle and the adrenal gland. This blockade inhibits the vasoconstrictive and aldosterone-secreting effects of angiotensin II, leading to vasodilation, decreased blood pressure, and reduced sodium and water retention.
Pharmacokinetics
Onset
Antihypertensive effect begins within 2 hours, maximum effect within 2-4 hours.
Excretion
Excreted primarily via the feces (80%) and urine (20%) as unchanged drug.
Half life
Elimination half-life is approximately 6-9 hours.
Absorption
Rapidly absorbed after oral administration, with peak plasma concentrations reached within 1-2 hours. Bioavailability is approximately 30-40%.
Metabolism
Undergoes minimal hepatic metabolism, primarily through non-enzymatic cleavage.
Side Effects
Contraindications
- Hypersensitivity to Abazin or any excipients
- Pregnancy (second and third trimesters)
- Concomitant use with aliskiren in patients with diabetes or renal impairment (GFR <60 mL/min/1.73 m²)
Drug Interactions
Lithium
Increased serum lithium concentrations and toxicity.
Aliskiren
Concomitant use is contraindicated in patients with diabetes or renal impairment due to increased risk of hyperkalemia, hypotension, and renal impairment.
NSAIDs (Non-Steroidal Anti-inflammatory Drugs)
May reduce the antihypertensive effect and increase the risk of renal function deterioration, especially in elderly or dehydrated patients.
Potassium-sparing diuretics/Potassium supplements
May lead to hyperkalemia (e.g., spironolactone, amiloride).
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Limited data are available on overdose in humans. The most likely manifestations of overdose would be hypotension and tachycardia; bradycardia could occur from parasympathetic stimulation. Treatment should be symptomatic and supportive.
Pregnancy & Lactation
Abazino is contraindicated during the second and third trimesters of pregnancy due to risks of fetal toxicity and death. It is not recommended during the first trimester. It is unknown whether Abazin is excreted in human milk, so caution should be exercised when administered to a nursing mother.
