Acefenac 100 mg Tablet contains Aceclofenac, a non-steroidal anti-inflammatory drug (NSAID), used for pain relief and reduction of inflammation in various conditions such as osteoarthritis, rheumatoid arthritis, and ankylosing spondylitis.
Uses & Indications
Dosage
Adults
The recommended dose is 100 mg twice daily, one tablet in the morning and one in the evening.
Elderly
No dosage adjustment is generally required, but caution should be exercised due to increased risk of side effects; the lowest effective dose should be used.
Renal_impairment
No dosage adjustment is usually necessary for mild renal impairment, but caution is advised. Avoid use in severe renal impairment.
How to Take
Acefenac 100 mg tablets should be swallowed whole with sufficient liquid, preferably with or after food, to minimize gastrointestinal discomfort.
Mechanism of Action
Aceclofenac works by inhibiting the enzyme cyclooxygenase (COX), which is responsible for the synthesis of prostaglandins. Prostaglandins are mediators of inflammation, pain, and fever. By reducing prostaglandin production, Aceclofenac exerts its anti-inflammatory, analgesic, and antipyretic effects.

Extensively metabolized in the liver, primarily by CYP2C9, into its main metabolite 4'-hydroxyaceclofenac.
Side Effects
Contraindications
- Hypersensitivity to Aceclofenac or any other NSAID
- Active gastrointestinal bleeding or peptic ulcer
- Severe heart failure, hepatic or renal impairment
- Third trimester of pregnancy
- History of asthma, urticaria, or allergic-type reactions after taking aspirin or other NSAIDs
Drug Interactions
Corticosteroids
Increased risk of gastrointestinal ulceration or bleeding.
Lithium and Methotrexate
Increased plasma levels of lithium and methotrexate.
Cyclosporine and Tacrolimus
Increased risk of nephrotoxicity.

Overdose
Symptoms of overdose may include nausea, vomiting, epigastric pain, gastrointestinal bleeding, dizziness, headache, and drowsiness. In severe cases, renal failure and liver damage may occur. Management is primarily supportive and symptomatic; gastric lavage or activated charcoal may be considered soon after ingestion.
Pregnancy & Lactation
Aceclofenac is contraindicated in the third trimester of pregnancy due to potential effects on fetal cardiovascular system. It should be avoided during early pregnancy and lactation unless the potential benefits outweigh the risks. Consult a doctor.
