Overview of the medicine
Acemox 250 mg Tablet contains Amoxicillin, a penicillin-type antibiotic used to treat a wide variety of bacterial infections. It works by stopping the growth of bacteria. This antibiotic treats only bacterial infections and will not work for viral infections (such as common cold, flu).
Uses & Indications
Dosage
Adults
Typical dosage is 250 mg to 500 mg every 8 hours, or 500 mg to 875 mg every 12 hours, depending on the severity and type of infection. Max dose: 4.5 g/day.
Elderly
No specific dose adjustment is usually required unless there is significant renal impairment. Dosage should be based on renal function.
Renal_impairment
Dosage reduction may be necessary in patients with severe renal impairment (e.g., CrCl < 30 mL/min). Consult physician for specific guidelines.
How to Take
Take the tablet orally with a glass of water, with or without food. It is generally recommended to take it with food if gastrointestinal upset occurs. Do not crush or chew the tablet; swallow it whole.
Mechanism of Action
Amoxicillin is a bactericidal antibiotic that acts by inhibiting bacterial cell wall synthesis. It prevents cross-linking of peptidoglycan polymers, which are a major component of bacterial cell walls. This leads to cell lysis and bacterial death.
Pharmacokinetics
Onset
Within 30-60 minutes after oral administration.
Excretion
Primarily excreted unchanged by the kidneys (glomerular filtration and tubular secretion) into the urine. About 60% of an oral dose is excreted unchanged within 6 hours.
Half life
Approximately 1-1.5 hours in adults with normal renal function.
Absorption
Rapidly and well absorbed from the gastrointestinal tract, especially when taken orally. Peak plasma concentrations are usually achieved within 1-2 hours.
Metabolism
Partially metabolized in the liver to inactive metabolites; about 10-25% of the dose is metabolized.
Side Effects
Contraindications
- •Hypersensitivity to amoxicillin, other penicillins, or any component of the formulation.
- •History of severe allergic reaction (e.g., anaphylaxis) to any beta-lactam antibiotic (e.g., cephalosporins, carbapenems).
Drug Interactions
Warfarin
May increase the anticoagulant effect of warfarin, requiring monitoring of INR.
Allopurinol
Increased risk of skin rash when co-administered with allopurinol.
Methotrexate
May increase methotrexate levels, leading to increased toxicity.
Oral Contraceptives
May reduce the effectiveness of oral contraceptives; advise using alternative birth control methods.
Storage
Store in a cool, dry place, below 30°C. Protect from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose may include gastrointestinal symptoms such as nausea, vomiting, and diarrhea. In rare cases, renal impairment, crystalluria, or convulsions may occur. Treatment is symptomatic and supportive. Gastric lavage or activated charcoal may be considered soon after ingestion.
Pregnancy & Lactation
Pregnancy Category B. Amoxicillin is generally considered safe for use during pregnancy; however, use only if clearly needed and prescribed by a doctor. Small amounts are excreted into breast milk; generally considered compatible with breastfeeding, but monitor the infant for diarrhea or rash.
