Description
Overview of the medicine
Acliz 50 mg Chewable Tablet is an antihistamine used to relieve symptoms associated with seasonal and perennial allergic rhinitis, such as sneezing, runny nose, itchy nose or throat, and watery eyes. It is also effective in treating symptoms of chronic idiopathic urticaria.
Uses & Indications
Dosage
Adults
One 50 mg chewable tablet once daily, preferably in the evening. May be increased to 100 mg daily in divided doses if required.
Elderly
No specific dose adjustment is usually required unless there is significant renal or hepatic impairment. Start with the lowest effective dose.
Renal_impairment
For moderate to severe renal impairment (CrCl <50 mL/min), dose should be reduced to 25 mg once daily or 50 mg every other day.
How to Take
Chew the tablet thoroughly before swallowing. Can be taken with or without food. Do not swallow whole.
Mechanism of Action
Acliz selectively inhibits peripheral H1-receptors, thereby preventing histamine from binding to these receptors. This action blocks the effects of histamine, such as vasodilation, increased capillary permeability, and itching, which are responsible for allergic symptoms.
Pharmacokinetics
Onset
Within 30-60 minutes.
Excretion
Excreted mainly through urine (both unchanged drug and metabolites) and feces.
Half life
Approximately 8-12 hours.
Absorption
Rapidly absorbed after oral administration, reaching peak plasma concentrations within 1-2 hours.
Metabolism
Primarily metabolized in the liver, likely via CYP450 enzymes.
Side Effects
Contraindications
- Hypersensitivity to Acliz or any of its components
- Children under 6 years of age (for chewable form)
Drug Interactions
Ketoconazole, Erythromycin
May increase plasma concentrations of Acliz due to inhibition of its metabolism.
CNS Depressants (e.g., alcohol, sedatives)
May enhance CNS depressant effects, leading to increased drowsiness.
Storage
Store in a cool, dry place, below 30°C. Protect from light and moisture. Keep out of reach of children.
Overdose
Symptoms may include drowsiness, tachycardia, headache, and anticholinergic effects. Treatment is symptomatic and supportive. Gastric lavage or activated charcoal may be considered if recent ingestion. Hemodialysis is not effective.
Pregnancy & Lactation
Pregnancy Category B. Use only if clearly needed during pregnancy. Excreted in breast milk; caution advised during lactation, as it may cause drowsiness in nursing infants.
