Adagel Plus 0.1%/2.5% Gel is a combination topical medication used for the treatment of acne vulgaris. It combines Adapalene, a retinoid-like compound, with Benzoyl Peroxide, an antibacterial and keratolytic agent.
Uses & Indications
Dosage
Adults
Apply a thin layer of gel to all affected areas of the face and/or trunk once daily in the evening after washing. Avoid contact with eyes, lips, nostrils, and mucous membranes.
Elderly
No specific dose adjustment is required for elderly patients, as systemic absorption is minimal.
Renal_impairment
No specific dose adjustment is required for patients with renal impairment due to minimal systemic absorption.
How to Take
For topical dermatological use only. Cleanse the skin thoroughly before applying the gel. Apply a thin film to the entire affected area (not just individual lesions) once daily in the evening. Wash hands thoroughly after application.
Mechanism of Action
Adapalene normalizes follicular keratinization and has anti-inflammatory properties, which helps to prevent the formation of new acne lesions. Benzoyl Peroxide is an antimicrobial that reduces the number of Propionibacterium acnes (P. acnes) bacteria on the skin and also acts as a keratolytic agent, helping to shed dead skin cells and unclog pores.
Pharmacokinetics
Onset
Visible improvement typically observed within 1 to 4 weeks, with full therapeutic effect often seen after 8 to 12 weeks of continuous use.
Excretion
Systemically absorbed adapalene and its metabolites are excreted primarily via the biliary route. Benzoic acid, the metabolite of benzoyl peroxide, is excreted in the urine.
Half life
Systemic half-life is not clinically relevant for topical administration due to minimal absorption. Benzoyl peroxide is rapidly metabolized to benzoic acid in the skin.
Absorption
Both adapalene and benzoyl peroxide have minimal systemic absorption when applied topically, leading to very low plasma concentrations.
Metabolism
Systemically absorbed adapalene is primarily metabolized in the liver via O-demethylation, hydroxylation, and conjugation. Benzoyl peroxide is converted to benzoic acid within the skin.
