Overview of the medicine
Alfasin-XR 10 mg tablet contains Alfuzosin Hydrochloride, an alpha-1 adrenergic blocker, used to treat the symptoms of benign prostatic hyperplasia (BPH) such as difficulty in urination, weak stream, and frequent urination.
Uses & Indications
Dosage
Adults
One 10 mg extended-release tablet once daily, immediately after the same meal each day.
Elderly
No dosage adjustment needed for elderly patients based on age alone, but monitor blood pressure.
Renal_impairment
No dosage adjustment needed for mild to moderate renal impairment. Use with caution in severe renal impairment (creatinine clearance < 30 mL/min).
How to Take
Take orally, once daily, immediately after a meal. Swallow the tablet whole; do not crush, chew, or divide.
Mechanism of Action
Alfuzosin selectively blocks alpha-1 adrenergic receptors in the prostate, bladder neck, and prostatic urethra. This blockade causes relaxation of the smooth muscle in these areas, leading to increased urine flow and a reduction in BPH symptoms.
Pharmacokinetics
Onset
Symptomatic improvement may be observed within 1-2 weeks.
Excretion
Mainly through feces (75-91%), a smaller portion via urine (11%).
Half life
Approximately 8-10 hours for the extended-release formulation.
Absorption
Approximately 49% when taken after a meal. Food enhances absorption.
Metabolism
Extensive hepatic metabolism, primarily via CYP3A4.

Contraindications

Drug Interactions
Other alpha-blockers
Increased risk of hypotension.
Antihypertensive medications
Enhanced hypotensive effect, risk of orthostatic hypotension.
Erectile dysfunction drugs (e.g., sildenafil, tadalafil)
Increased risk of hypotension, use with caution and consider dose separation.
Potent CYP3A4 inhibitors (ketoconazole, itraconazole, ritonavir)
Significantly increases Alfuzosin plasma concentrations.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Overdose may lead to hypotension. Treatment should be supportive, including placing the patient in a supine position, and if necessary, administering vasopressors and intravenous fluids. Dialysis is unlikely to be beneficial.
Pregnancy & Lactation
Alfuzosin is not indicated for use in women and should not be used during pregnancy or lactation.
