Uses & Indications

How to Take

Mechanism of Action
Desloratadine is a potent, long-acting, selective peripheral histamine H1-receptor antagonist. It inhibits the release of histamine from mast cells and basophils, thereby suppressing the allergic reaction.
Pharmacokinetics
Onset
Onset of action typically occurs within 30 minutes to 1 hour.
Excretion
Excreted almost equally via urine and feces as metabolites.
Half life
Mean plasma elimination half-life is approximately 27 hours.
Absorption
Rapidly absorbed after oral administration. Peak plasma concentrations reached within approximately 3 hours.
Metabolism
Extensively metabolized, primarily to 3-hydroxydesloratadine, which is an active metabolite. Metabolism involves CYP3A4 (minor) and UGTs.
Side Effects
Contraindications
- Hypersensitivity to desloratadine, loratadine, or to any of the excipients.
Drug Interactions
Cimetidine
Increased plasma concentrations of desloratadine reported, but no clinically significant adverse effects.

Desloratadine is categorized as Pregnancy Category C. It should be used during pregnancy only if the potential benefit justifies the potential risk to the fetus. Desloratadine is excreted into breast milk; therefore, a decision should be made whether to discontinue nursing or to discontinue the drug, taking into account the importance of the drug to the mother.
