Amlodipine is a calcium channel blocker used to treat high blood pressure (hypertension) and chest pain (angina). It works by relaxing blood vessels, making it easier for the heart to pump blood.
Uses & Indications
Dosage
Adults
Initial dose is 5 mg orally once daily. Maximum dose is 10 mg once daily.
Elderly
A starting dose of 2.5 mg once daily may be considered. Standard adult dose if tolerated.
Renal_impairment
No dosage adjustment is required for patients with renal impairment.
Hepatic_impairment
A starting dose of 2.5 mg once daily is recommended in patients with hepatic impairment.
How to Take

Mechanism of Action
Amlodipine is a dihydropyridine calcium antagonist that inhibits the transmembrane influx of calcium ions into vascular smooth muscle and cardiac muscle. This causes vascular smooth muscle relaxation and peripheral vasodilation, leading to a reduction in blood pressure and myocardial oxygen demand.

Excreted primarily in urine as inactive metabolites (60%) and less than 10% as unchanged drug.

Clarithromycin

Storage

Overdose
Symptoms include excessive peripheral vasodilation with marked hypotension and possibly reflex tachycardia. Management involves active cardiovascular support, including monitoring cardiac and respiratory function, elevation of extremities, and monitoring circulating fluid volume and urine output. Intravenous calcium gluconate may be beneficial.
