Description
Overview of the medicine
Amocil 125 mg Suspension is an antibiotic from the penicillin group, used to treat a wide range of bacterial infections. It works by stopping the growth of bacteria.
Uses & Indications
Dosage
Adults
For specific adult infections requiring 125 mg, the typical dose is 125 mg orally every 8 hours. However, adults commonly use higher strengths (e.g., 250 mg or 500 mg capsules/tablets) for most infections. Pediatric dosage (for children < 40 kg): 20-40 mg/kg/day in divided doses every 8 hours, or 25-45 mg/kg/day in divided doses every 12 hours, depending on infection severity.
Elderly
Dosage considerations are similar to adults; renal function should be assessed and dose adjusted if impairment is present.
Renal_impairment
Dose adjustments are required for patients with significant renal impairment. For creatinine clearance 10-30 mL/min, 125-250 mg every 12 hours. For creatinine clearance < 10 mL/min, 125-250 mg every 24 hours. Pediatric renal dosing should be based on weight and severity.
How to Take
Shake well before use. Administer orally using a measuring spoon or oral syringe for accurate dosing. Can be taken with or without food.
Mechanism of Action
Amoxicillin works by inhibiting bacterial cell wall synthesis. It binds to penicillin-binding proteins (PBPs) located in the bacterial cell wall, leading to inhibition of peptidoglycan synthesis, which in turn causes bacterial cell lysis and death.
Pharmacokinetics
Onset
Approximately 1-2 hours for peak plasma concentrations.
Excretion
Primarily excreted unchanged by the kidneys via tubular secretion and glomerular filtration. About 60% of an oral dose is excreted unchanged in urine within 6-8 hours.
Half life
Approximately 1-1.5 hours in adults with normal renal function. Prolonged in renal impairment.
Absorption
Rapidly and well absorbed from the gastrointestinal tract, even in the presence of food. Bioavailability is approximately 75-90%.
Metabolism
Partially metabolized to inactive penicilloic acid.
Side Effects
Contraindications
- Hypersensitivity to amoxicillin, other penicillins, or any component of the formulation
- History of severe immediate hypersensitivity reaction (e.g., anaphylaxis) to any other beta-lactam antibacterial agent
Drug Interactions
Warfarin
May prolong prothrombin time; monitor INR closely.
Probenecid
Increases amoxicillin blood levels by decreasing renal tubular secretion.
Methotrexate
May increase methotrexate toxicity; monitor levels.
Oral Contraceptives
Amoxicillin may reduce the efficacy of oral contraceptives (rarely).
Storage
Store dry powder below 30°C, protected from light and moisture. Once reconstituted, store in a refrigerator (2-8°C) and discard after 7-14 days (check specific product label).
Overdose
Symptoms of overdose may include nausea, vomiting, diarrhea, and rarely, convulsions. Treatment is supportive and symptomatic. Gastric lavage or activated charcoal may be considered if recent. Hemodialysis can remove amoxicillin from the circulation.
Pregnancy & Lactation
Pregnancy Category B. Generally considered safe during pregnancy. Excreted in breast milk in small amounts; generally considered compatible with breastfeeding, but monitor infant for potential side effects (e.g., diarrhea, candidiasis).

YesClinical Trials
Extensive clinical data supports the efficacy and safety of amoxicillin for various bacterial infections, primarily established in trials conducted in the 1970s and 1980s. Ongoing surveillance studies confirm its continued utility.
Lab Monitoring
- Renal function tests (for prolonged therapy or renal impairment)
- Liver function tests (rarely, in patients with pre-existing hepatic impairment)
- Complete blood count (for prolonged therapy)
Doctor Notes
- Emphasize importance of completing full antibiotic course.
- Counsel patients on potential allergic reactions and advise seeking immediate medical attention if signs occur.
- Be aware of potential drug interactions, especially with anticoagulants and methotrexate.
Patient Guidelines
- Complete the full course of antibiotics as prescribed, even if symptoms improve.
- Do not save medicine for future infections or share it with others.
- Shake the suspension well before each use.
- Use the provided measuring device for accurate dosing.
Missed Dose Advice
Take the missed dose as soon as you remember. If it is almost time for your next dose, skip the missed dose and continue your regular dosing schedule. Do not double the dose.
Driving Precautions
Generally, amoxicillin does not impair the ability to drive or operate machinery. However, if you experience dizziness or other side effects, avoid these activities.
Lifestyle Advice
- Maintain good hygiene to prevent reinfection.
- Drink plenty of fluids to stay hydrated, especially if experiencing diarrhea.
- Avoid alcohol during antibiotic treatment as it may worsen side effects like nausea or dizziness.
