Description
Overview of the medicine
Amoxicillin is a broad-spectrum penicillin antibiotic used to treat a variety of bacterial infections.
Uses & Indications
Dosage
Adults
Typically 250 mg to 500 mg every 8 hours, or 500 mg to 875 mg every 12 hours, depending on infection severity. For suspensions, 125 mg/5 ml is primarily for pediatric use, so adult dosage would require higher strength preparations.
Elderly
Similar to adults, but dosage adjustment may be needed based on renal function.
Renal_impairment
Dosage adjustment is necessary for patients with impaired renal function (e.g., reduce dose or extend dosing interval).
How to Take
Shake well before use. Can be taken with or without food. Use a measuring spoon or cup for accurate dosing.
Mechanism of Action
Amoxicillin works by inhibiting the synthesis of bacterial cell walls, leading to bacterial lysis and death.
Pharmacokinetics
Onset
Approximately 30-60 minutes.
Excretion
Primarily excreted by the kidneys through both glomerular filtration and tubular secretion.
Half life
About 1 to 1.5 hours in adults with normal renal function.
Absorption
Rapidly and well absorbed from the gastrointestinal tract, especially when taken orally.
Metabolism
Partially metabolized to inactive metabolites; a significant portion is excreted unchanged.
Side Effects
Contraindications
- History of hypersensitivity to penicillin antibiotics
- History of hypersensitivity to cephalosporins (cross-reactivity)
Drug Interactions
Warfarin
May enhance anticoagulant effect, monitor INR.
Allopurinol
Increased risk of skin rash.
Methotrexate
May increase methotrexate toxicity.
Oral Contraceptives
May reduce efficacy of oral contraceptives.
Storage
Store dry powder below 30°C, protected from moisture. Store reconstituted suspension in a refrigerator (2-8°C) and do not freeze.
Overdose
Symptoms may include gastrointestinal distress (nausea, vomiting, diarrhea) and renal impairment (in rare cases). Treatment is symptomatic and supportive; hemodialysis may be used in severe cases.
Pregnancy & Lactation
Generally considered safe during pregnancy (Category B). Small amounts pass into breast milk, generally considered safe but use with caution and monitor infant for diarrhea or rash.
