Adapalene is a topical retinoid-like compound that normalizes the differentiation of follicular epithelial cells resulting in decreased microcomedone formation.
Uses & Indications
Dosage
Adults
Apply a thin film to affected areas once daily at bedtime.
Elderly
No specific dosage adjustments are necessary for elderly patients.
Renal_impairment
No dosage adjustments are necessary for patients with renal impairment.
How to Take
Apply a thin layer to clean, dry skin once daily at bedtime. Avoid contact with eyes, lips, and mucous membranes.
Mechanism of Action
Adapalene binds to specific retinoic acid nuclear receptors but does not bind to the cytosolic receptor protein. It reduces inflammation by inhibiting the migration of polymorphonuclear leukocytes.
Pharmacokinetics
Onset
Clinical improvement is typically observed after 8-12 weeks of treatment.
Excretion
Excretion is mainly via bile.
Half life
The half-life is approximately 10-25 hours in plasma.
Absorption
Adapalene is poorly absorbed through the skin. Systemic exposure is low.
Metabolism
Metabolism is primarily by O-demethylation, hydroxylation, and conjugation.

Contraindications

Drug Interactions
Photosensitizing Medications

Storage
Store at room temperature, away from heat and direct light.
Overdose
Excessive topical application may lead to marked redness, peeling, or discomfort. Treatment is symptomatic and supportive.
Pregnancy & Lactation
Pregnancy Category C. Use during pregnancy is not recommended. It is not known whether adapalene is excreted in breast milk. Caution should be exercised when administered to a nursing woman.
