Description
Overview of the medicine
Aribex is a hypothetical new generation analgesic targeting acute to moderate pain. It is currently under advanced clinical trials for safety and efficacy.
Uses & Indications
Dosage
Adults
50 mg orally, every 4-6 hours as needed. Maximum 200 mg per day.
Elderly
Consider lower starting dose (e.g., 25 mg) and adjust based on renal function and tolerability.
Renal_impairment
Use with caution. Dosage reduction recommended for severe impairment (CrCl < 30 mL/min).
How to Take
Take orally with a full glass of water, with or without food. If gastric upset occurs, take with food or milk.
Mechanism of Action
Aribex is believed to exert its analgesic effects by selectively inhibiting the cyclooxygenase-3 (COX-3) enzyme in the central nervous system, thereby reducing prostaglandin synthesis involved in pain signaling. It has minimal effect on COX-1 and COX-2 enzymes, potentially leading to fewer gastrointestinal and renal side effects.
Pharmacokinetics
Onset
Pain relief typically begins within 30 minutes of oral administration.
Excretion
Excreted mainly through urine (70% as metabolites, 10% as unchanged drug) and faeces (20%).
Half life
Approximately 4-6 hours.
Absorption
Rapidly absorbed from the gastrointestinal tract, peak plasma concentration reached within 1-2 hours.
Metabolism
Primarily metabolized in the liver via glucuronidation and hydroxylation pathways.
Side Effects
Contraindications
- Hypersensitivity to Aribex or any of its excipients
- Severe hepatic impairment
- Active peptic ulcer disease
Drug Interactions
SSRIs
Potential increased risk of gastrointestinal bleeding when co-administered.
Warfarin
May increase the risk of bleeding. Monitor INR closely.
Storage
Store below 30°C in a dry place, away from direct sunlight and moisture. Keep out of reach of children.
Overdose
Symptoms may include nausea, vomiting, abdominal pain, drowsiness, and in severe cases, liver or kidney damage. Management is supportive and symptomatic, including gastric lavage or activated charcoal if recent ingestion.
Pregnancy & Lactation
Category C in pregnancy (hypothetical). Use only if potential benefit justifies the potential risk to the fetus. Excreted in breast milk; advise caution in lactating mothers.
