Description
Overview of the medicine
Arodex 1 mg Tablet contains Anastrozole, an aromatase inhibitor used primarily in the treatment of hormone receptor-positive early breast cancer in postmenopausal women. It works by reducing the amount of estrogen produced by the body, thereby inhibiting the growth of estrogen-dependent tumors.
Uses & Indications
Dosage
Adults
One 1 mg tablet orally once daily.
Elderly
No dose adjustment required for elderly patients.
Renal_impairment
No dose adjustment required for mild to moderate renal impairment. Use with caution in severe renal impairment (creatinine clearance <30 mL/min).
How to Take
Oral. Can be taken with or without food. Should be taken at approximately the same time each day.
Mechanism of Action
Anastrozole is a potent, selective, non-steroidal aromatase inhibitor. It inhibits aromatase, the enzyme responsible for converting androgens into estrogens (estrone and estradiol) in peripheral tissues. By reducing estrogen levels, it inhibits the growth of estrogen-sensitive breast cancer cells.
Pharmacokinetics
Onset
Significant estrogen suppression observed within 24 hours of first dose.
Excretion
Mainly excreted via urine, with about 10% as unchanged drug.
Half life
Approximately 40-50 hours.
Absorption
Rapidly absorbed from the gastrointestinal tract; peak plasma concentrations reached within 2 hours.
Metabolism
Extensively metabolized in the liver, primarily via N-dealkylation and glucuronidation.
Side Effects
Contraindications
- Pre-menopausal women.
- Pregnancy and lactation.
- Known hypersensitivity to anastrozole or any component of the tablet.
Drug Interactions
Tamoxifen
Concurrent use with tamoxifen should be avoided as it may reduce anastrozole's plasma concentrations and efficacy.
Estrogen-containing therapies
Should not be co-administered as they would negate the pharmacological action of anastrozole.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
No specific antidote for anastrozole overdose. Treatment should be symptomatic and supportive, including general supportive care and frequent monitoring of vital signs. Emesis may be induced if the patient is alert.
Pregnancy & Lactation
Contraindicated in pregnant and lactating women. Women of childbearing potential should use effective contraception during treatment and for a period after stopping treatment.
