Description
Overview of the medicine
Artemet is a combination antimalarial drug used to treat uncomplicated malaria caused by Plasmodium falciparum.
Uses & Indications
Dosage
Adults
4 tablets as initial dose, followed by 4 tablets after 8 hours, then 4 tablets twice daily for the following 2 days (total 24 tablets). Must be taken with fatty food.
Elderly
Same as adults, provided normal renal and hepatic function.
Renal_impairment
No dosage adjustment is generally needed. However, monitor patients with severe renal impairment.
How to Take
Take with food, preferably fatty food to enhance absorption.
Mechanism of Action
Artemether is rapidly metabolized into dihydroartemisinin (DHA) which disrupts parasite electron transport and causes parasite death. Lumefantrine inhibits parasitic nucleic acid and protein synthesis.

Onset
Within hours of administration.
Excretion
Primarily excreted in the feces.
Half life
Artemether: 1-3 hours, Lumefantrine: 3-6 days
Absorption
Artemether absorption is rapid. Lumefantrine absorption is enhanced by food.
Metabolism
Artemether is metabolized in the liver by CYP3A4/5. Lumefantrine is metabolized by CYP3A4.

Contraindications

Drug Interactions
CYP3A4 inducers (e.g., rifampin)
May decrease artemether and lumefantrine levels.
CYP3A4 inhibitors (e.g., ketoconazole)
May increase lumefantrine levels.
Storage
Store below 30°C in a dry place, protected from light.
Overdose
In case of overdose, seek immediate medical attention. Symptoms may include nausea, vomiting, and cardiac abnormalities.
Pregnancy & Lactation
Use during pregnancy only if the potential benefit justifies the potential risk to the fetus. Consult a doctor if breastfeeding.
