Description
Overview of the medicine
Baclomark 10 mg Tablet contains Baclofen, a skeletal muscle relaxant used to treat spasticity resulting from multiple sclerosis, spinal cord injuries, or other spinal cord diseases. It helps reduce muscle stiffness, spasms, and pain, improving mobility.
Uses & Indications
Dosage
Adults
Initially, 5 mg three times daily. Dose should be increased gradually (e.g., by 5 mg increments every 3 days) up to a maximum of 20 mg three times daily, or as directed by physician.
Elderly
Lower initial doses and slower titration may be required. Generally, 5 mg twice daily, adjusted as needed.
Renal_impairment
Dosage should be reduced significantly, e.g., 5 mg once daily, and titrated carefully under medical supervision due to prolonged excretion.
How to Take
Oral administration. Should be taken with food or milk to minimize gastrointestinal irritation.
Mechanism of Action
Baclofen acts as a GABA-B receptor agonist, leading to hyperpolarization of neurons and reduced release of excitatory neurotransmitters. This reduces the excitability of afferent nerve fibers, inhibiting both monosynaptic and polysynaptic reflexes at the spinal cord level, thereby reducing muscle spasticity.
Pharmacokinetics
Onset
Within 1-2 hours
Excretion
Primarily excreted unchanged by the kidneys (70-80%) via urine within 72 hours.
Half life
3-4 hours
Absorption
Rapidly and extensively absorbed from the gastrointestinal tract. Peak plasma concentrations are achieved within 2-4 hours.
Metabolism
Minimally metabolized; approximately 15% is metabolized in the liver to inactive metabolites.
Side Effects
Contraindications
- •Hypersensitivity to Baclofen or any component of the formulation
- •Epilepsy (use with caution, may lower seizure threshold)
Drug Interactions
Antihypertensives
Increased hypotensive effects.

Increased CNS depression, drowsiness, and respiratory depression.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Symptoms include drowsiness, coma, respiratory depression, seizures, and hypotonia. Treatment is primarily supportive, focusing on maintaining airway and cardiovascular function. Gastric lavage may be considered.
