Description
Overview of the medicine
Bevacizumab is a recombinant humanized monoclonal antibody that selectively binds to vascular endothelial growth factor (VEGF), preventing its interaction with receptors on endothelial cells. This inhibits angiogenesis (the formation of new blood vessels), thereby slowing tumor growth and metastasis.
Uses & Indications
Dosage
Adults
Dosage varies by indication and body weight, typically 5 mg/kg, 7.5 mg/kg, 10 mg/kg, or 15 mg/kg administered intravenously every 2 or 3 weeks. Always follow specific oncology treatment protocols.
Elderly
No specific dose adjustments for elderly patients are generally required based on age alone, but careful monitoring for adverse effects is recommended.
Renal_impairment
No specific dose adjustment is required for patients with renal impairment.
How to Take
Administer intravenously as an infusion over 30 to 90 minutes. Do not administer as an intravenous push or bolus. The first infusion should be given over 90 minutes. If well tolerated, the second infusion may be given over 60 minutes. Subsequent infusions may be given over 30 minutes if the 60-minute infusion is well tolerated.
Mechanism of Action
Bevacizumab binds to and neutralizes VEGF, a key mediator of angiogenesis. By inhibiting VEGF, the formation of new blood vessels required for tumor growth and survival is suppressed. This leads to decreased tumor vascularization, reduced blood supply to the tumor, and ultimately, inhibition of tumor growth and metastasis.

Side Effects
Contraindications

- •Recent major surgery or unhealed wounds.
Drug Interactions
Sunitinib
Increased risk of microangiopathic hemolytic anemia, severe proteinuria, and hypertension.
Anthracyclines
Potential for increased cardiac dysfunction, though data is limited.
Radiation Therapy
Potential for increased risk of fistula formation, especially in patients with pelvic malignancies.
Platinum/Taxane-based Chemotherapy
