Description
Overview of the medicine
Caffo 500 mg Tablet contains Paracetamol, which is used to relieve mild to moderate pain and reduce fever. It works by blocking the production of certain chemical messengers that cause pain and fever.
Uses & Indications
Dosage
Adults
500-1000 mg (1-2 tablets) every 4-6 hours as needed, not to exceed 4000 mg (8 tablets) in 24 hours.
Elderly
Similar to adults; caution advised in patients with compromised hepatic or renal function.
Renal_impairment
Dose interval may need to be extended in severe renal impairment (e.g., 500 mg every 8-12 hours).
How to Take
Administer orally with water, with or without food. Do not exceed the recommended dose or duration.
Mechanism of Action
Paracetamol primarily inhibits prostaglandin synthesis in the central nervous system (CNS), resulting in analgesic (pain relief) and antipyretic (fever reduction) effects. It has weak peripheral anti-inflammatory activity.
Pharmacokinetics
Onset
Analgesic effect begins within 30-60 minutes; antipyretic effect within 30 minutes.
Excretion
Mainly excreted in urine as glucuronide and sulfate conjugates; less than 5% is excreted unchanged.
Half life
Plasma half-life is 1 to 4 hours, prolonged in hepatic dysfunction.
Absorption
Rapidly and almost completely absorbed from the gastrointestinal tract, with peak plasma concentrations occurring within 30-60 minutes.
Metabolism
Primarily metabolized in the liver via glucuronidation and sulfation, with a small amount by cytochrome P450 pathway.
Side Effects
Contraindications
- Hypersensitivity to Paracetamol or any component of the formulation.
- Severe hepatic impairment or active liver disease.
Drug Interactions
Alcohol
Increases the risk of hepatotoxicity, especially with chronic excessive alcohol consumption.
Warfarin
May increase the anticoagulant effect of warfarin, increasing bleeding risk.
Cholestyramine
Reduces paracetamol absorption if taken within one hour.
Other hepatotoxic drugs
Concurrent use may increase the risk of liver damage.
Metoclopramide/Domperidone
Increases paracetamol absorption rate.
Storage
Store below 30°C in a dry place, away from direct sunlight and moisture. Keep out of reach of children.
Overdose
Overdose can cause severe, potentially fatal liver damage. Symptoms include nausea, vomiting, abdominal pain. Management involves gastric lavage, activated charcoal, and administration of N-acetylcysteine (NAC) as an antidote.
Pregnancy & Lactation
Generally considered safe for use during pregnancy and lactation at recommended therapeutic doses. However, always consult a doctor before use.
