Overview of the medicine
Cinagen-XG 05-04 Eye Drop is an ophthalmic solution containing Olopatadine Hydrochloride and Ketorolac Tromethamine, indicated for the treatment of allergic conjunctivitis and associated ocular inflammation.
Uses & Indications
Dosage
Adults
Instill 1 drop in the affected eye(s) twice daily (approximately 8 hours apart), or as directed by a physician.
Elderly
No specific dosage adjustment is required for elderly patients.
Renal_impairment
No dosage adjustment is required for patients with renal impairment for ophthalmic use.
How to Take
For ophthalmic use only. Tilt head back, pull down the lower eyelid to form a pouch, and instill the drop into the conjunctival sac. Avoid touching the dropper tip to the eye or any other surface.
Mechanism of Action
Olopatadine acts as a selective H1-receptor antagonist and mast cell stabilizer, reducing histamine-mediated allergic reactions. Ketorolac inhibits prostaglandin synthesis by blocking cyclooxygenase (COX-1 and COX-2) enzymes, thereby reducing inflammation and pain.

Onset
Olopatadine: within minutes for itching relief. Ketorolac: within hours for anti-inflammatory effect.
Excretion
Primarily renal excretion for both active ingredients (systemic).
Half life
Olopatadine plasma half-life is approximately 3 hours (systemic). Ketorolac plasma half-life is approximately 4-6 hours (systemic, negligible for ophthalmic use).
Absorption
Minimal systemic absorption following ocular administration. Acts locally in the eye.
Metabolism
Olopatadine: primarily hepatic, minimal metabolism. Ketorolac: extensively metabolized in the liver, primarily via hydroxylation and conjugation.

- Hypersensitivity to Olopatadine, Ketorolac, or any excipients in the formulation.
- Patients with known allergic reactions to other NSAIDs.
