Description
Overview of the medicine
Citicoline is a brain chemical that occurs naturally in the body. It is used as a medicine to improve brain function, particularly in conditions like stroke, head injury, and cognitive impairment due to age or disease. It helps in the repair of damaged cell membranes and increases the levels of important neurotransmitters.
Uses & Indications
Dosage
Adults
500-2000 mg daily, typically divided into two doses. For acute stroke, 500 mg twice daily. For cognitive impairment, 500-1000 mg daily.
Elderly
No specific dose adjustment required, usually same as adults.
Renal_impairment
No specific dose adjustment necessary as excretion is not primarily renal.
How to Take
Citicoline tablets can be taken with or without food. Swallow the tablet whole with a glass of water. Do not crush or chew.
Mechanism of Action
Citicoline is converted to cytidine and choline in the body. Choline is then used in the synthesis of acetylcholine (a neurotransmitter) and phosphatidylcholine (a component of cell membranes). It enhances phospholipid synthesis in neuronal membranes, repairs damaged cells, and has neuroprotective effects by stabilizing cell membranes and reducing free radical damage.
Pharmacokinetics
Onset
Effects may be noticed within hours for acute conditions, but cognitive benefits are usually seen after several weeks of continuous use.
Excretion
Primarily excreted as CO2 in the breath (for choline) and via urine (for cytidine and its metabolites).
Half life
The plasma half-life of choline is about 1-4 hours, while cytidine has a longer half-life after being incorporated into various cellular components.
Absorption
Well absorbed orally (>90%). Rapidly hydrolyzed in the gut wall and liver to choline and cytidine.
Metabolism
Metabolized in the liver and gut wall. Choline is incorporated into phospholipids and acetylcholine. Cytidine is incorporated into nucleic acids or catabolized.
Side Effects
Contraindications
- Hypersensitivity to citicoline or any component of the formulation.
- Conditions involving persistent hypertonia (increased muscle tone).
Drug Interactions
Levodopa
May enhance the effects of Levodopa (used in Parkinson's disease).
Meclofenoxate
Should not be administered concomitantly with meclofenoxate.
Centrophenoxine
Should not be administered concomitantly with centrophenoxine.
Storage
Store in a cool, dry place below 30°C. Protect from light and moisture. Keep out of reach of children.
Overdose
Citicoline has low toxicity. In case of accidental overdose, symptomatic and supportive treatment should be given.
Pregnancy & Lactation
