Description
Overview of the medicine
Cloradex eye drop is a combination medication containing an antibiotic (Chloramphenicol) and a corticosteroid (Dexamethasone). It is used to treat bacterial eye infections where inflammation is also present.
Uses & Indications
Dosage
Adults
1-2 drops into the affected eye(s) 3-4 times daily or as directed by the physician.
Elderly
No specific dosage adjustment required, use with caution.
Renal_impairment
No dosage adjustment needed due to minimal systemic absorption.
How to Take
Wash hands before use. Tilt head back, pull down lower eyelid to form a pouch. Instill drops without touching the eye with the dropper tip. Close eye gently for 1-2 minutes. Do not rinse the dropper.
Mechanism of Action
Chloramphenicol inhibits bacterial protein synthesis by binding to the 50S ribosomal subunit. Dexamethasone is a potent synthetic corticosteroid that suppresses inflammation by inhibiting various inflammatory mediators.
Pharmacokinetics
Onset
Anti-inflammatory effects of Dexamethasone may begin within hours. Antibiotic effects of Chloramphenicol within 24-48 hours.
Excretion
Locally metabolized and excreted via normal ocular fluid turnover. Minimal systemic excretion.
Half life
Not significant for ocular application due to minimal systemic absorption.
Absorption
Minimal systemic absorption occurs after ocular administration. Local absorption into ocular tissues.
Metabolism
Primarily local metabolism in ocular tissues; systemic metabolism (hepatic) for absorbed components is negligible.
Side Effects
Contraindications
- Hypersensitivity to Chloramphenicol, Dexamethasone, or any component of the formulation
- Viral or fungal eye infections
- Untreated parasitic infections
- Mycobacterial infections of the eye
- Glaucoma or ocular hypertension (unless specifically prescribed and monitored)
Drug Interactions
Immunosuppressants
May increase the risk of systemic effects of Dexamethasone with prolonged use.
Other ophthalmic preparations
Administer with an interval of at least 5-10 minutes.
Storage
Store in a cool, dry place, below 30°C. Protect from light. Do not freeze. Keep out of reach of children.
