Description
Overview of the medicine
Coralben-DS 600 mg Tablet contains Albendazole, an anthelmintic medication used to treat a variety of parasitic worm infections. It is effective against various intestinal and tissue-dwelling parasites.
Uses & Indications
Dosage
Adults
Dosage varies significantly based on the type of parasitic infection. For most intestinal worm infections, a single dose of 400 mg is common. For systemic infections like neurocysticercosis or hydatid disease, the dosage can be 400 mg twice daily for varying durations (e.g., 8-30 days), or as directed by a physician. A 600 mg tablet may be used for specific regimens or severe cases as determined by a specialist.
Elderly
No specific dose adjustment is generally required for elderly patients, but caution should be exercised due to potential age-related decrease in organ function.
Renal_impairment
No dose adjustment is generally required in renal impairment, as albendazole and its primary metabolite are minimally excreted by the kidneys. However, caution is advised.
How to Take
Take orally with food, preferably a fatty meal, to enhance absorption. Tablets can be chewed, crushed, or swallowed whole depending on patient preference and specific instructions.
Mechanism of Action
Albendazole selectively binds to beta-tubulin, inhibiting microtubule polymerization in parasitic helminths. This leads to impaired glucose uptake and depletion of glycogen stores in the worm, ultimately causing paralysis and death of the parasite.
Pharmacokinetics
Onset
Onset of anthelmintic action varies depending on the parasite and location; clinical resolution may take days to weeks.
Excretion
Primarily excreted in the urine as metabolites, with a small amount excreted in bile.
Half life
Albendazole sulfoxide (active metabolite): 8-12 hours.
Absorption
Poorly absorbed orally (<5%). Absorption is enhanced with fatty meals. Rapidly and extensively metabolized in the liver to albendazole sulfoxide, which is the primary active metabolite.
Metabolism
Extensively metabolized in the liver via first-pass metabolism to its primary active metabolite, albendazole sulfoxide, which is then further metabolized.
Side Effects
Contraindications
- •Known hypersensitivity to albendazole or any component of the formulation
- •Pregnancy (especially in the first trimester, unless benefits outweigh risks)
- •Bone marrow suppression (pre-existing)
Drug Interactions
Theophylline
Albendazole may decrease the plasma concentrations of Theophylline, requiring monitoring of theophylline levels.
Cimetidine, Dexamethasone, Praziquantel
Cimetidine, Dexamethasone, and Praziquantel may increase the plasma concentrations of albendazole sulfoxide, potentially increasing side effects.
Storage
Store in a cool, dry place below 30°C, away from direct light and moisture. Keep out of reach of children.
Overdose
In case of overdose, symptomatic and supportive treatment should be initiated. Gastric lavage and activated charcoal may be considered if ingestion is recent.
Pregnancy & Lactation
Pregnancy Category C. Should not be used during the first trimester of pregnancy unless the potential benefits outweigh the risks. In later trimesters, use only if clearly needed. It is unknown if albendazole or its metabolites are excreted in human milk; use with caution during lactation.
