Description
Overview of the medicine
Defungi 50 mg capsule contains Fluconazole, an antifungal agent used to treat various fungal infections, including candidiasis and cryptococcal meningitis.
Uses & Indications
Dosage
Adults
Varies based on infection type and severity. For vaginal candidiasis, a single 150 mg oral dose. For oropharyngeal candidiasis, 50-100 mg once daily for 7-14 days. For systemic infections, 400 mg on day 1 followed by 200-400 mg once daily.
Elderly
Dosage adjustment not usually required unless renal function is impaired.
Renal_impairment
Dosage adjustment required. For creatinine clearance <50 mL/min, reduce dose by 50%.
How to Take
Capsules can be taken with or without food. Swallow whole with water.
Mechanism of Action
Fluconazole inhibits fungal cytochrome P450-dependent 14-alpha-demethylase, which is essential for ergosterol biosynthesis. This inhibition leads to the accumulation of 14-alpha-methyl sterols and a decrease in ergosterol, disrupting fungal cell membrane integrity.

Excretion
Primarily renal excretion (approximately 80% as unchanged drug).

Absorption
Well absorbed orally, bioavailability >90%.
