Description
Overview of the medicine
Deloxi contains Desloratadine, a long-acting tricyclic antihistamine with selective H1-receptor antagonist activity. It is used to relieve symptoms of allergic rhinitis and chronic idiopathic urticaria.
Uses & Indications
Dosage
Adults
Adults and adolescents 12 years of age and over: 5 mg once daily.
Elderly
No dosage adjustment generally required for elderly patients.
Renal_impairment
For patients with renal or hepatic impairment, a starting dose of 5 mg every other day is recommended.
How to Take
May be taken with or without food. Swallow the tablet whole with water.
Mechanism of Action
Desloratadine is a non-sedating, long-acting antihistamine with potent, selective peripheral H1-receptor antagonist activity. It inhibits the release of histamine from mast cells and basophils, thereby reducing allergic symptoms.

Onset
Within 30 minutes to 1 hour.
Excretion
Excreted almost equally in urine and feces.
Half life
Approximately 27 hours.
Absorption
Rapidly absorbed after oral administration. Peak plasma concentrations are reached in approximately 3 hours.
Metabolism
Extensively metabolized to 3-hydroxydesloratadine, an active metabolite, primarily by CYP2C8 and CYP2D6.
Side Effects
Contraindications
- Hypersensitivity to Desloratadine, Loratadine, or any component of the formulation.
- Patients with severe renal impairment.
Drug Interactions
Cimetidine
Increased plasma concentrations of Desloratadine.
Fluoxetine
Increased plasma concentrations of Desloratadine.
Erythromycin
Increased plasma concentrations of Desloratadine.
Ketoconazole
Increased plasma concentrations of Desloratadine.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose may include somnolence, increased heart rate, and headache. In the event of overdose, symptomatic and supportive treatment is recommended. Desloratadine is not removed by hemodialysis.
Pregnancy & Lactation
Pregnancy: Category C. Use only if potential benefit justifies the potential risk to the fetus. Lactation: Desloratadine is excreted into breast milk. A decision should be made whether to discontinue nursing or to discontinue the drug.
