Description
Overview of the medicine
Denamine 4 mg Tablet is an antihistamine used to relieve symptoms of allergic reactions such as sneezing, runny nose, watery eyes, and itching. It works by blocking the action of histamine, a natural substance that causes allergic symptoms.
Uses & Indications
Dosage
Adults
4 mg every 4-6 hours, not exceeding 24 mg in 24 hours.
Elderly
Lower doses may be required (e.g., 2 mg every 4-6 hours) due to increased sensitivity and risk of side effects.
Renal_impairment
Use with caution; dose adjustment may be necessary in severe renal impairment.
How to Take
Take orally with or without food. Swallow the tablet whole with water.
Mechanism of Action
Chlorpheniramine Maleate acts as a competitive antagonist of histamine at H1 receptors, thereby preventing the actions of histamine, such as vasodilation, increased capillary permeability, and spasm of smooth muscle.
Pharmacokinetics
Onset
30-60 minutes
Excretion
Mainly excreted in urine as metabolites and some unchanged drug.
Half life
12-43 hours (highly variable)
Absorption
Well absorbed from the gastrointestinal tract, with peak plasma concentrations occurring within 2-6 hours.
Metabolism
Extensively metabolized in the liver by demethylation and hydroxylation.
Side Effects
Contraindications
- Hypersensitivity to chlorpheniramine or any component of the formulation.
- Acute asthma attacks.
- Newborns or premature infants.
- Concomitant use with MAO inhibitors.
Drug Interactions
Monoamine Oxidase Inhibitors (MAOIs)
Prolongation and intensification of anticholinergic effects and CNS depression.
Anticholinergic Drugs (e.g., tricyclic antidepressants)
Additive anticholinergic effects (dry mouth, blurred vision, urinary retention).
CNS Depressants (e.g., alcohol, benzodiazepines, opioids)
Increased sedative effect.
Storage
Store in a cool, dry place below 30°C, away from direct sunlight and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose include drowsiness, CNS depression or stimulation (especially in children), dry mouth, flushed skin, dilated pupils, fever, and gastrointestinal symptoms. Severe overdose can lead to convulsions, coma, and respiratory depression. Treatment is symptomatic and supportive, including gastric lavage or activated charcoal.
Pregnancy & Lactation
Use in pregnancy only if the potential benefit outweighs the potential risk to the fetus. Not recommended during breastfeeding as it may pass into breast milk and cause drowsiness in the infant. Consult a doctor.
