Description
Overview of the medicine
Desloratadine is a non-sedating antihistamine used to relieve symptoms associated with allergic rhinitis (including perennial allergic rhinitis) and chronic idiopathic urticaria in children.
Uses & Indications
Dosage
Adults
For children 6-11 months: 1 mg (2 mL) once daily; 1-5 years: 1.25 mg (2.5 mL) once daily; 6-11 years: 2.5 mg (5 mL) once daily.
Elderly
Not applicable for pediatric drops. Consult physician for adult formulations.
Renal_impairment
For patients with renal impairment, a lower starting dose (e.g., 0.5 mg every other day for children 6-11 months) or less frequent administration may be considered. Consult a physician.
How to Take
For oral administration. Can be taken with or without food. Measure the dose accurately using the provided dropper or measuring spoon.
Mechanism of Action
Desloratadine is a potent, selective, peripheral H1-receptor antagonist. It inhibits histamine release from mast cells and basophils, and also inhibits other allergic inflammatory mediators.

Onset
Symptomatic relief usually occurs within 30 minutes to 1 hour.
Excretion
Excreted primarily via urine and feces as conjugated metabolites.
Half life
The elimination half-life is approximately 27 hours.
Absorption
Rapidly absorbed after oral administration, with peak plasma concentrations reached in approximately 3 hours.
Metabolism
Extensively metabolized to 3-hydroxydesloratadine, an active metabolite, primarily by CYP2C8 and CYP3A4.

Cimetidine
Concomitant administration with cimetidine resulted in increased plasma concentrations of desloratadine.
