Description
Overview of the medicine
Desloratadine is a non-drowsy antihistamine used to relieve symptoms associated with allergic rhinitis (including seasonal and perennial) and chronic idiopathic urticaria.
Uses & Indications
Dosage
Adults
5 mg once daily.
Elderly
No specific dose adjustment is generally required, but caution is advised in patients with renal or hepatic impairment.
Renal_impairment
For severe renal impairment (creatinine clearance <30 mL/min), 5 mg every other day.
How to Take
Take orally, with or without food. Swallow the tablet whole with water.
Mechanism of Action
Desloratadine is a potent, long-acting, selective peripheral H1-receptor antagonist. It inhibits histamine release and other pro-inflammatory mediators from mast cells and basophils, thereby reducing allergy symptoms.
Pharmacokinetics
Onset
Within 30 minutes of administration.
Excretion
Excreted equally in urine and feces as metabolic products.
Half life
Approximately 27 hours.
Absorption
Well absorbed orally; peak plasma concentrations reached approximately 3 hours after administration.
Metabolism
Extensively metabolized, primarily via hydroxylation followed by conjugation. Metabolites are pharmacologically active but contribute less to overall activity.
Side Effects
Contraindications
- Known hypersensitivity to desloratadine, loratadine, or any component of the formulation.
Drug Interactions

Concomitant use may increase plasma concentrations of desloratadine.
Fluoxetine
Concomitant use may increase plasma concentrations of desloratadine.
Azithromycin

Storage
Store below 30°C in a dry place, away from direct sunlight and moisture. Keep out of reach of children.

Doctor Notes
- Emphasize that desloratadine is a non-sedating antihistamine, offering relief without significant drowsiness for most patients.
- Advise patients on appropriate dosing for renal impairment, especially for severe cases.
- Counsel patients on potential drug interactions, though major interactions are uncommon.
Patient Guidelines
- Do not exceed the recommended dose.
- Inform your doctor or pharmacist about all medications, supplements, and herbal products you are taking.
- If you experience any severe or persistent side effects, contact your doctor immediately.
Missed Dose Advice
If you miss a dose, take it as soon as you remember. If it is almost time for your next dose, skip the missed dose and resume your regular dosing schedule. Do not take a double dose to make up for a missed one.
Driving Precautions
Desloratadine generally does not affect the ability to drive or operate machinery. However, if you experience drowsiness or dizziness, avoid these activities.
Lifestyle Advice
- Although Deslosil is generally non-drowsy, alcohol consumption should be avoided or minimized as it may enhance central nervous system depressant effects.
