Description
Overview of the medicine
Desloratadine is a non-drowsy antihistamine used to relieve symptoms associated with allergic rhinitis (including seasonal and perennial) and chronic idiopathic urticaria.
Uses & Indications
Dosage
Adults
5 mg once daily.
Elderly
No specific dose adjustment is generally required, but caution is advised in patients with renal or hepatic impairment.
Renal_impairment
For severe renal impairment (creatinine clearance <30 mL/min), 5 mg every other day.
How to Take
Take orally, with or without food. Swallow the tablet whole with water.
Mechanism of Action
Desloratadine is a potent, long-acting, selective peripheral H1-receptor antagonist. It inhibits histamine release and other pro-inflammatory mediators from mast cells and basophils, thereby reducing allergy symptoms.

Onset
Within 30 minutes of administration.
Excretion
Excreted equally in urine and feces as metabolic products.
Half life
Approximately 27 hours.
Absorption
Well absorbed orally; peak plasma concentrations reached approximately 3 hours after administration.
Metabolism
Extensively metabolized, primarily via hydroxylation followed by conjugation. Metabolites are pharmacologically active but contribute less to overall activity.

Contraindications
- •Known hypersensitivity to desloratadine, loratadine, or any component of the formulation.
Drug Interactions
Cimetidine
Concomitant use may increase plasma concentrations of desloratadine.
Fluoxetine
Concomitant use may increase plasma concentrations of desloratadine.
Azithromycin
No clinically significant pharmacokinetic interaction observed.
Erythromycin
Concomitant use may increase plasma concentrations of desloratadine.
Ketoconazole
Concomitant use may increase plasma concentrations of desloratadine.
Storage
Store below 30°C in a dry place, away from direct sunlight and moisture. Keep out of reach of children.
Overdose
In case of overdose, seek emergency medical attention. Treatment should be symptomatic and supportive.
Pregnancy & Lactation
Use during pregnancy only if clearly needed and the potential benefit outweighs the potential risk to the fetus. It is not recommended during lactation as desloratadine is excreted into breast milk.
