Description
Overview of the medicine
Dexzole 30 mg capsule contains Dexlansoprazole, a proton pump inhibitor (PPI) used to reduce stomach acid production. It is primarily used for the treatment of gastroesophageal reflux disease (GERD) and healing of erosive esophagitis.
Uses & Indications
Dosage
Adults
For healing EE: 60 mg once daily for up to 8 weeks. For maintenance of EE and relief of heartburn: 30 mg once daily for up to 6 months. For symptomatic non-erosive GERD: 30 mg once daily for 4 weeks.
Elderly
No dosage adjustment is necessary for elderly patients.
Renal_impairment
No dosage adjustment is necessary for patients with renal impairment.
How to Take
Take the capsule whole, with or without food. Do not chew or crush. For patients who have difficulty swallowing, the capsule can be opened and the intact granules sprinkled onto a tablespoon of applesauce and swallowed immediately.
Mechanism of Action
Dexlansoprazole is a proton pump inhibitor that suppresses gastric acid secretion by specific inhibition of the H+/K+-ATPase in the gastric parietal cell. By acting specifically on the proton pump, dexlansoprazole blocks the final step of acid production.
Pharmacokinetics
Onset
Acid suppression typically begins within hours of the first dose.
Excretion
Excreted in both urine (approximately 50%) and feces (approximately 47%) as inactive metabolites.
Half life
Terminal elimination half-life is approximately 1-2 hours.
Absorption
Rapidly absorbed after oral administration. It has a dual delayed-release formulation resulting in two peaks in plasma concentration (1-2 hours and 4-5 hours post-dose).
Metabolism
Extensively metabolized in the liver, primarily by cytochrome P450 (CYP) enzymes CYP2C19 and CYP3A4.
Side Effects
Contraindications
- Hypersensitivity to dexlansoprazole or any component of the formulation.
- Concomitant use with rilpivirine-containing products.
Drug Interactions
Warfarin
Increased INR and prothrombin time.
Clopidogrel
Potential reduction in antiplatelet activity.
Methotrexate
Increased and prolonged elevation of methotrexate and/or its metabolite serum levels.
Atazanavir, Nelfinavir
Reduced absorption, potentially reducing antiviral efficacy.
Iron Salts, Ketoconazole, Itraconazole
Reduced absorption of drugs that depend on gastric pH for absorption.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
No specific antidote. In case of overdose, discontinue the drug and treat symptomatically and provide supportive care. Dexlansoprazole is not dialyzable.
