Overview of the medicine
Diacon 80 mg tablet contains Gliclazide, an oral hypoglycaemic agent used to control high blood sugar levels in patients with type 2 diabetes mellitus. It stimulates insulin release from the pancreatic beta cells.
Uses & Indications
Dosage
Adults
Initial dose 40-80 mg daily, increasing gradually to 320 mg daily if needed. Doses >160 mg should be given in two divided doses.
Elderly
Similar to adults, but initial doses should be lower (e.g., 40 mg daily) and titrated carefully due to increased risk of hypoglycemia.
Renal_impairment
No dosage adjustment needed for mild to moderate renal impairment. Use with caution in severe impairment; consider lower doses and close monitoring.
How to Take
Take orally, preferably with breakfast or the first main meal of the day. Swallow whole with water, do not crush or chew.
Mechanism of Action
Gliclazide primarily acts by stimulating insulin secretion from the pancreatic β-cells. It binds to sulfonylurea receptors on the β-cell membrane, leading to the closure of ATP-sensitive potassium channels, depolarization of the membrane, and influx of calcium ions. This influx triggers the release of insulin.

Miconazole
Potentiates the hypoglycemic effect, leading to severe hypoglycemia. Concomitant use is contraindicated.
Anticoagulants (e.g., Warfarin)
Gliclazide may potentiate the effect of anticoagulants, requiring dose adjustment of the anticoagulant.
Phenylbutazone, Alcohol, Sulfonamides, Fluoroquinolones
May enhance the blood glucose lowering effect, increasing risk of hypoglycemia.
Corticosteroids, Danazol, Chlorpromazine, Beta-2 agonists (oral), Ritodrine, Salbutamol (IV), Terbutaline (IV)
May reduce the hypoglycemic effect, leading to increased blood glucose levels.
