Overview of the medicine
Diatrol MR is an oral antidiabetic medicine containing Gliclazide Modified Release. It is used to lower blood glucose levels in adults with type 2 diabetes mellitus.
Uses & Indications
Dosage
Adults
Initial dose: 30 mg once daily, taken with breakfast. May be increased to 60 mg, 90 mg, or 120 mg daily, according to blood glucose response, with increments of at least 1 month interval. Maximum recommended dose is 120 mg daily.
Elderly
Similar to adult dosage, but caution is advised due to increased risk of hypoglycemia. Start with a lower dose.
Renal_impairment
In mild to moderate renal impairment, no dose adjustment is usually required. In severe renal impairment, it is generally not recommended.
How to Take
Take the tablet whole with breakfast, once daily. Do not chew or crush the tablet.
Mechanism of Action
Gliclazide primarily stimulates insulin secretion from pancreatic beta cells. It also enhances peripheral insulin sensitivity and reduces hepatic glucose production.

Gradual, due to modified release.
Excretion
Primarily excreted in urine (60-70%) and feces (10-20%) as metabolites.

Approximately 12-20 hours.

Contraindications
- Type 1 diabetes mellitus
- Diabetic ketoacidosis
- Severe renal or hepatic insufficiency
- Hypersensitivity to gliclazide or other sulfonylureas
- Pregnancy and lactation
- Concomitant treatment with miconazole.
Drug Interactions
Miconazole (oral gel, systemic)
Potentiates hypoglycemic effect, potentially leading to coma.
Corticosteroids, danazol, chlorpromazine, diuretics (thiazides), oral contraceptives
May reduce the hypoglycemic effect (hyperglycemia).
Phenylbutazone, alcohol, other antidiabetics, beta-blockers, fluconazole, H2-antagonists, MAOIs, sulfonamides, NSAIDs
May potentiate the hypoglycemic effect.
Storage
Store below 30°C, in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Overdose may lead to severe hypoglycemia, which can be treated by administering glucose (oral or intravenous). In severe cases, hospitalization may be required.
Pregnancy & Lactation
Gliclazide is contraindicated during pregnancy and breastfeeding due to potential fetal harm and excretion into breast milk. Insulin is preferred for glycemic control during pregnancy.
