Overview of the medicine
Doxilin 100 mg Capsule contains Doxycycline, which is a broad-spectrum tetracycline antibiotic used to treat various bacterial infections including respiratory tract infections, urinary tract infections, skin infections, sexually transmitted infections, rickettsial infections, and malaria prophylaxis.
Uses & Indications
Dosage
Adults
Typical dose is 100 mg once or twice daily, depending on the infection. For severe infections, 100 mg twice daily. For acne, 50 mg daily. For malaria prophylaxis, 100 mg once daily starting 1-2 days before travel and continuing for 4 weeks after leaving the endemic area.
Elderly
No specific dose adjustment is generally required, but caution is advised due to potential comorbidities.
Renal_impairment
No dose adjustment is usually necessary for patients with renal impairment, as doxycycline is primarily excreted non-renally.
How to Take
Take orally with a full glass of water while sitting upright to prevent esophageal irritation or ulceration. It can be taken with food or milk if stomach upset occurs, although dairy products may slightly reduce absorption (take 1-2 hours before or 4-6 hours after). Do not lie down for at least 30 minutes after taking the medicine.
Mechanism of Action
Doxycycline inhibits bacterial protein synthesis by reversibly binding to the 30S ribosomal subunit of susceptible bacteria. This prevents the attachment of aminoacyl tRNA to the acceptor site on the mRNA-ribosome complex, thereby inhibiting peptide chain elongation and bacterial growth.

Onset
Variable, generally within hours of administration.
Excretion
Primarily excreted unchanged in feces via intestinal secretion, with a small amount excreted renally.
Half life
Approximately 16-24 hours.
Absorption
Well absorbed orally (90-100%). Absorption is minimally affected by food or dairy products.
Metabolism
Minimally metabolized in the liver; no significant first-pass metabolism.
Side Effects
Contraindications
- •Hypersensitivity to doxycycline or other tetracyclines

- •Children under 8 years of age (risk of permanent tooth discoloration and enamel hypoplasia)
Drug Interactions
Penicillins
Tetracyclines may interfere with the bactericidal action of penicillins. Avoid concomitant use.
Oral contraceptives
May reduce the effectiveness of oral contraceptives. Advise alternative contraception.
Warfarin and other anticoagulants
Doxycycline may potentiate the anticoagulant effects. Close monitoring of INR/PT is recommended.
Barbiturates, Carbamazepine, Phenytoin
May reduce the half-life of doxycycline due to enzyme induction.
Antacids (containing aluminum, calcium, magnesium), Iron preparations, Bismuth subsalicylate
Can decrease the absorption of doxycycline. Administer 1-2 hours before or 4-6 hours after doxycycline.
Storage
Store below 30°C in a dry place, away from direct sunlight and moisture. Keep out of reach of children.
Overdose
In case of overdose, symptoms may include nausea, vomiting, and diarrhea. Management typically involves gastric lavage, followed by symptomatic and supportive treatment. Dialysis is not effective in removing doxycycline from the blood.
Pregnancy & Lactation
Doxycycline is classified as Pregnancy Category D, indicating evidence of fetal risk. It should not be used during pregnancy as it can cause permanent tooth discoloration and inhibit bone growth in the fetus. It is also excreted in breast milk and is not recommended during lactation due to potential adverse effects on the nursing infant.
