Domperidone is a dopamine antagonist that helps relieve nausea, vomiting, and certain digestive issues by increasing gastrointestinal motility. This 30 mg tablet is typically used for sustained release or specific dosage regimens.
Uses & Indications
Dosage
Adults
Typically 30 mg once daily, preferably 15-30 minutes before a meal. For standard domperidone (10mg), the dose is 10-20 mg three times daily. Max 30 mg/day.
Elderly
Similar to adults, but use with caution due to potential for cardiac effects.
Renal_impairment
In severe renal impairment (creatinine clearance <30 ml/min), the frequency of dosing should be reduced to once or twice daily depending on severity (e.g., 10 mg once or twice daily).
How to Take
Take orally, preferably 15-30 minutes before meals. Do not crush or chew sustained-release tablets.
Mechanism of Action
It selectively blocks dopamine D2 receptors in the chemoreceptor trigger zone (CTZ) in the brain, reducing nausea and vomiting. Additionally, it acts as a prokinetic agent by enhancing gastrointestinal motility, increasing lower esophageal sphincter pressure, and accelerating gastric emptying.

- Prolactin-releasing pituitary tumor (prolactinoma)
- Moderate to severe hepatic impairment
- Known existing prolongation of cardiac QT intervals or significant cardiac disease
- Concomitant use of potent CYP3A4 inhibitors (e.g., ketoconazole, erythromycin)
- Gastrointestinal hemorrhage, mechanical obstruction, or perforation
