Description
Overview of the medicine
Efl oxin 250 mg Suspension is an oral antibacterial medication containing Ofloxacin, a fluoroquinolone antibiotic. It is used to treat a wide range of bacterial infections in children and adults, including respiratory, urinary tract, skin, and soft tissue infections. It works by stopping the growth of bacteria.
Uses & Indications
Dosage
Adults
While suspension is primarily for pediatric use, adults requiring liquid formulation may take 200-400 mg twice daily, as directed by a physician. Dosage varies based on infection type and severity.
Elderly
Dose adjustment may be necessary based on renal function, often starting at the lower end of the dosage range.
Children
Typically 10-15 mg/kg/day, divided into two doses, not exceeding 400 mg/day. For Efl oxin 250 mg Suspension, a common dose might be 5-10 mL twice daily, depending on weight and infection. Always follow pediatrician's advice.
Renal_impairment
Dosage adjustment is required for patients with creatinine clearance <50 mL/min. For example, the first dose may be normal, followed by half the recommended dose every 24 hours.
How to Take
For oral use. Shake the bottle well before each use. Can be taken with or without food. It is crucial to complete the entire prescribed course of medication, even if symptoms improve, to prevent the development of antibiotic resistance.
Mechanism of Action
Ofloxacin inhibits bacterial DNA gyrase and topoisomerase IV, enzymes essential for bacterial DNA replication, transcription, repair, and recombination. By blocking these enzymes, Ofloxacin prevents bacteria from multiplying, leading to their death.
Pharmacokinetics
Onset
Therapeutic effects typically begin within a few hours to days, depending on the type and severity of infection.
Excretion
Primarily excreted unchanged in the urine (70-80% within 24-48 hours). A small amount is excreted in feces.
Half life
Approximately 5 to 8 hours in individuals with normal renal function.
Absorption
Rapid and well-absorbed from the gastrointestinal tract after oral administration. Peak plasma concentrations are usually achieved within 1 to 2 hours.
Metabolism
Minimally metabolized in the liver, with less than 10% of the administered dose undergoing biotransformation.
Side Effects
Contraindications
- •Hypersensitivity to Ofloxacin, other fluoroquinolones, or any component of the formulation.
- •History of tendon disorders related to fluoroquinolone use.
- •Children and adolescents in the growth phase, due to the risk of cartilage damage (though used in specific pediatric infections when benefits outweigh risks).
- •Patients with epilepsy or a history of seizure disorders (use with caution).
- •Pregnancy and lactation.
Drug Interactions
Warfarin
Ofloxacin may enhance the anticoagulant effect of warfarin, increasing the risk of bleeding. Close monitoring of INR is recommended.
Theophylline
Concomitant use may increase serum concentrations of theophylline, leading to increased risk of theophylline toxicity. Monitor theophylline levels.
Corticosteroids
Increased risk of tendinitis and tendon rupture when co-administered with corticosteroids.
NSAIDs (Non-steroidal anti-inflammatory drugs)
Co-administration with NSAIDs may increase the risk of central nervous system stimulation and convulsions.
Antacids (containing magnesium, aluminum), Iron, Zinc supplements
Concomitant administration may significantly decrease Ofloxacin absorption. Administer Ofloxacin at least 2 hours before or 2 hours after these agents.
Storage
Store below 30°C in a dry place, protected from light. Do not freeze. Keep out of reach of children.
Overdose
In case of overdose, symptoms may include central nervous system effects (e.g., confusion, dizziness, seizures), gastrointestinal disturbances (e.g., nausea, vomiting). Management should be symptomatic and supportive. Gastric lavage and activated charcoal may be considered. Adequate hydration should be maintained.
Pregnancy & Lactation
Pregnancy Category C. Ofloxacin should be used during pregnancy only if the potential benefit justifies the potential risk to the fetus. It is excreted in human milk; therefore, due to the potential for serious adverse reactions in nursing infants, a decision should be made whether to discontinue nursing or to discontinue the drug, taking into account the importance of the drug to the mother.
