Description
Overview of the medicine
Efrad 125 mg Suspension is an antibiotic from the cephalosporin class, used to treat a wide range of bacterial infections. It works by stopping the growth of bacteria.
Uses & Indications
Dosage
Adults
Typically, adult doses of Cefradine are higher (e.g., 250-500 mg every 6 hours), usually administered as capsules or tablets. The 125 mg suspension is primarily for pediatric use. For children (over 1 month): 25-50 mg/kg/day in 2-4 divided doses. For severe infections, up to 100 mg/kg/day. A typical dose for 125 mg/5 ml suspension might be 5 ml (125 mg) 2-4 times daily, depending on age and weight.
Elderly
For elderly patients with normal renal function, no specific dosage adjustment is required. Monitor renal function and adjust dosage if impairment is present.
Renal_impairment
For patients with impaired renal function, dosage adjustment is necessary. For mild to moderate impairment, the dosage interval may be extended. For severe impairment, dose reduction or further extension of interval may be needed as per physician's advice and creatinine clearance levels.
How to Take
Shake the bottle well before each use. Administer orally with or without food. Use a measuring spoon or oral syringe for accurate dosing.
Mechanism of Action
Cefradine is a first-generation cephalosporin antibiotic that inhibits bacterial cell wall synthesis by binding to penicillin-binding proteins (PBPs), leading to bacterial cell lysis and death.
Pharmacokinetics
Onset
Within 30-60 minutes
Excretion
Primarily excreted unchanged by the kidneys via glomerular filtration and tubular secretion
Half life
Approximately 0.5-1.5 hours in adults with normal renal function
Absorption
Rapidly and well absorbed from the gastrointestinal tract, especially after oral administration. Peak plasma concentrations are achieved within 1-2 hours.
Metabolism
Minimally metabolized
Side Effects
Contraindications
- Hypersensitivity to cefradine or other cephalosporins
- History of severe hypersensitivity reaction to any beta-lactam antibiotic (e.g., penicillins)
Drug Interactions
Probenecid
Increases plasma concentrations and prolongs the half-life of cefradine by reducing renal excretion.
Oral Anticoagulants
May increase the anticoagulant effect, requiring closer monitoring of INR.
Storage
Store in a cool, dry place below 30°C, protected from light and moisture. Keep out of reach of children. After reconstitution, store in a refrigerator (2-8°C) and use within 7 days.
Overdose
Symptoms of overdose may include nausea, vomiting, diarrhea, and in severe cases, neurological disturbances (e.g., seizures). Treatment is primarily supportive. Hemodialysis may be useful in reducing serum cefradine concentrations.
Pregnancy & Lactation
Pregnancy Category B. Studies in animals have shown no evidence of harm to the fetus. Use in pregnant women should be done with caution and only if clearly needed. Excreted in breast milk in small amounts; use with caution in nursing mothers.

YesClinical Trials
Cefradine has been extensively studied in clinical trials for its efficacy and safety in various bacterial infections. Most trials established its role as a broad-spectrum cephalosporin.
Lab Monitoring
- Renal function tests (e.g., creatinine, BUN) in patients with pre-existing renal impairment or during prolonged therapy.
- Complete blood count (CBC) during prolonged therapy.
- Liver function tests (LFTs) if clinically indicated.
Doctor Notes
- Assess patient history for hypersensitivity reactions to beta-lactam antibiotics.
- Adjust dose in renal impairment. Monitor for signs of superinfection.
- Emphasize completing the full course to prevent resistance.
Patient Guidelines
- Complete the full course of treatment as prescribed by your doctor, even if symptoms improve.
- Do not save leftover medicine for future infections.
- Report any severe or persistent side effects to your doctor.
Missed Dose Advice
If you miss a dose, take it as soon as you remember. If it is almost time for your next dose, skip the missed dose and resume your regular dosing schedule. Do not double the dose to catch up.
