Overview of the medicine
Etrib (Etoricoxib) is a non-steroidal anti-inflammatory drug (NSAID) used to relieve pain and inflammation in conditions like osteoarthritis, rheumatoid arthritis, ankylosing spondylitis, and gout.
Uses & Indications
Dosage
Adults
Osteoarthritis: 60 mg once daily. Rheumatoid Arthritis, Ankylosing Spondylitis: 90 mg once daily. Acute Gout: 120 mg once daily (max 8 days). Acute Pain/Dysmenorrhea: 120 mg once daily (max 8 days) or 90 mg once daily for up to 3 days for dental surgery pain.
Elderly
No dosage adjustment generally required, but caution is advised due to potential age-related decrease in renal function.
Renal_impairment
Mild to moderate renal impairment (CrCl 30-80 mL/min): No dosage adjustment required. Severe renal impairment (CrCl <30 mL/min): Not recommended.
How to Take
Take orally, with or without food. Swallow the tablet whole with a glass of water.
Mechanism of Action
Etoricoxib selectively inhibits the cyclooxygenase-2 (COX-2) enzyme, which is responsible for the synthesis of prostaglandins, mediators of pain and inflammation. By inhibiting COX-2, it reduces the production of these inflammatory substances.

Analgesic effects typically begin within 24 minutes.
Excretion
Primarily excreted via the kidneys (70%) and in feces (20%) as metabolites.
Half life
Approximately 22 hours.
Absorption
Rapidly absorbed from the gastrointestinal tract, with an oral bioavailability of approximately 100%. Peak plasma concentrations are reached in about 1 hour.
Metabolism
Extensively metabolized in the liver, primarily via the cytochrome P450 (CYP) isoenzyme CYP3A4, forming inactive metabolites.
Side Effects
Contraindications
- Hypersensitivity to etoricoxib or any component of the product
- History of asthma, urticaria, or allergic-type reactions after taking aspirin or other NSAIDs
- Congestive heart failure (NYHA II-IV)
- Established ischemic heart disease, peripheral arterial disease, and/or cerebrovascular disease
- Uncontrolled hypertension (blood pressure consistently above 140/90 mmHg)
- Active peptic ulceration or gastrointestinal bleeding
- Severe hepatic dysfunction (Child-Pugh score >9)
- Severe renal dysfunction (CrCl <30 mL/min)

Overdose
In case of overdose, seek immediate medical attention. Management is supportive and symptomatic; gastric decontamination (e.g., activated charcoal) may be considered if ingestion is recent.
Pregnancy & Lactation
Not recommended during the third trimester of pregnancy due to potential for premature closure of the fetal ductus arteriosus. Use with caution during lactation; it is unknown whether etoricoxib is excreted in human milk.
