Overview of the medicine
Eurofil (Sildenafil Citrate) is a medication primarily used to treat erectile dysfunction (impotence) in men and pulmonary arterial hypertension. It works by increasing blood flow to the penis during sexual stimulation, leading to an erection. For pulmonary hypertension, it relaxes blood vessels in the lungs.
Uses & Indications
Dosage
Adults
For ED: Usually 50 mg orally, as needed, approximately 1 hour before sexual activity. Can be adjusted to 25 mg or 100 mg. Max 1 dose per day. For PAH: 20 mg three times daily.
Elderly
Consider starting with 25 mg for ED due to potential for decreased clearance.
Renal_impairment
For severe renal impairment (creatinine clearance <30 mL/min), consider starting with 25 mg for ED.
How to Take
Take orally, with or without food. For ED, take approximately 1 hour before sexual activity. For PAH, take regularly as prescribed.
Mechanism of Action
Sildenafil is a selective inhibitor of cyclic guanosine monophosphate (cGMP)-specific phosphodiesterase type 5 (PDE5). In erectile dysfunction, it enhances the effects of nitric oxide (NO) released during sexual stimulation, leading to increased cGMP levels in the corpus cavernosum, resulting in smooth muscle relaxation and inflow of blood. In pulmonary hypertension, it relaxes the smooth muscle cells in the pulmonary vascular bed.
