Description
Overview of the medicine
Ezevent is a medication used to lower high cholesterol levels in the blood. It works by inhibiting the absorption of cholesterol in the small intestine, thereby reducing the amount of cholesterol available to the liver, which leads to a reduction in LDL ('bad') cholesterol.
Uses & Indications
Dosage
Adults
The recommended dose is 10 mg once daily, taken with or without food.
Elderly
No dose adjustment is required for elderly patients.
Renal_impairment
No dose adjustment is necessary for patients with mild to moderate renal impairment. Caution is advised in severe renal impairment (CrCl <30 mL/min).
How to Take
Ezevent should be taken orally once daily, with or without food. It can be taken at any time of day.
Mechanism of Action
Ezetimibe selectively inhibits the absorption of dietary and biliary cholesterol by the small intestine, without affecting the absorption of fat-soluble vitamins, triglycerides, or bile acids. It binds to the Niemann-Pick C1-like 1 (NPC1L1) protein, which is crucial for cholesterol uptake.
Pharmacokinetics
Onset
Lipid-lowering effects are observed within 2 weeks.
Excretion
Mainly excreted in the feces (approximately 78%) and to a lesser extent in the urine (approximately 11%).
Half life
Approximately 22 hours for both ezetimibe and its active metabolite.
Absorption
Ezetimibe is rapidly absorbed and extensively metabolized to its active phenolic glucuronide form (ezetimibe-glucuronide) in the small intestine and liver.
Metabolism
Primarily in the small intestine and liver via glucuronide conjugation (Phase II reaction).
Side Effects
Contraindications
- Hypersensitivity to the active substance or to any of the excipients.
- When Ezevent is co-administered with a statin, the combination is contraindicated in patients with active liver disease or unexplained persistent elevations in serum transaminases.
Drug Interactions
Statins
Increased risk of myopathy/rhabdomyolysis when co-administered with statins.
Fibrates
Coadministration with fibrates (especially gemfibrozil) increases the risk of cholelithiasis and cholecystitis. Not recommended with gemfibrozil.
Cyclosporine
Concomitant administration with cyclosporine may increase ezetimibe concentrations. Monitor cyclosporine levels.
Cholestyramine
May decrease the absorption of ezetimibe. Take Ezevent at least 2 hours before or 4 hours after cholestyramine.
Storage
Store in a cool, dry place, below 30°C. Protect from light and moisture. Keep out of reach of children.
Overdose
In the event of an overdose, symptomatic and supportive measures should be taken. No specific antidote is available.
Pregnancy & Lactation
Pregnancy Category C. Use during pregnancy only if the potential benefit justifies the potential risk to the fetus. It is not known whether ezetimibe is excreted in human milk; caution should be exercised when administered to a nursing woman.
