Description
Overview of the medicine
Ezevent 10 mg Tablet contains Ezetimibe, a cholesterol absorption inhibitor. It is used to lower high cholesterol levels in the blood, either alone or in combination with statins. It works by reducing the absorption of cholesterol from the small intestine.
Uses & Indications
Dosage
Adults
The recommended dose is 10 mg once daily, with or without food.
Elderly
No dosage adjustment required for elderly patients.
Renal_impairment
No dosage adjustment required for mild to moderate renal impairment. Caution advised in severe renal impairment.
How to Take
Take orally, once daily, with or without food, at any time of the day. Do not crush or chew the tablet. If taken with a statin, it can be taken at the same time. If taken with a bile acid sequestrant, take Ezetimibe at least 2 hours before or 4 hours after the sequestrant.
Mechanism of Action
Ezetimibe selectively inhibits the absorption of dietary and biliary cholesterol by the small intestine, without affecting the absorption of fat-soluble vitamins, triglycerides, or bile acids. It targets the Niemann-Pick C1-Like 1 (NPC1L1) protein, which is involved in cholesterol uptake.
Pharmacokinetics
Onset
Lipid-lowering effects typically observed within 1-2 weeks.
Excretion
Mainly excreted in feces (approximately 78%) and urine (approximately 11%).
Half life
Approximately 22 hours for total ezetimibe (ezetimibe and ezetimibe-glucuronide).
Absorption
Well absorbed orally, undergoes extensive enterohepatic recirculation. Peak plasma concentrations (Cmax) are reached within 4-12 hours.
Metabolism
Primarily metabolized in the small intestine and liver via glucuronide conjugation, followed by biliary excretion.
Side Effects
Contraindications
- •Hypersensitivity to Ezetimibe or any component of the formulation.
- •When co-administered with a statin, refer to the statin's contraindications.
- •Active liver disease or unexplained persistent elevations of serum transaminases (when co-administered with a statin).
Drug Interactions
Cyclosporine
May increase ezetimibe exposure. Monitor cyclosporine levels and ezetimibe effects.
Fibrates (e.g., Gemfibrozil)
May increase the risk of cholelithiasis and enhance the concentration of ezetimibe. Co-administration is not recommended.
Warfarin, other coumarin anticoagulants
Monitor INR when initiating ezetimibe.
Bile Acid Sequestrants (e.g., Cholestyramine)
May decrease the absorption of ezetimibe. Take ezetimibe at least 2 hours before or 4 hours after these agents.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
In the event of an overdose, symptomatic and supportive measures should be employed. Ezetimibe is not dialyzable.
Pregnancy & Lactation
Pregnancy Category C. Use during pregnancy only if the potential benefit justifies the potential risk to the fetus. It is not known whether ezetimibe is excreted in human milk. Exercise caution when administered to a nursing woman.
