Description
Overview of the medicine
Femara (Letrozole) is an aromatase inhibitor used in the treatment of hormone-receptor-positive breast cancer in postmenopausal women. It works by reducing the amount of estrogen the body makes.
Uses & Indications
Dosage
Adults
2.5 mg orally once daily.
Elderly
No dose adjustment is required in elderly patients.
Renal_impairment
No dose adjustment is required for patients with mild to moderate renal impairment. Data for severe renal impairment is limited.
How to Take
Femara tablets should be taken orally, once daily, with or without food. It is recommended to take the tablet at approximately the same time each day.
Mechanism of Action
Letrozole selectively inhibits aromatase, an enzyme that converts androgens to estrogens. This reduction in estrogen levels can inhibit the growth of hormone-dependent breast cancer cells.

Onset
Estrogen suppression begins within 24 hours.
Excretion
Primarily renal (about 90% of the administered dose), with about 6% excreted unchanged in urine.
Half life
Approximately 2 days (48 hours).
Absorption
Rapidly and completely absorbed from the gastrointestinal tract.
Metabolism
Hepatic metabolism by cytochrome P450 isoenzymes CYP2A6 and CYP3A4 to an inactive carbinol metabolite.
Side Effects
Contraindications

- •Premenopausal women, pregnancy, and lactation.
Drug Interactions
Tamoxifen
Concomitant use with tamoxifen should be avoided as it may reduce letrozole's efficacy.
Estrogen-containing medications
Avoid concomitant use as they may counteract the pharmacological action of letrozole.
Storage
Store at room temperature (20°C-25°C), away from moisture and heat. Keep out of reach of children.
Overdose
There is no specific antidote for Femara overdose. Treatment should be symptomatic and supportive.
Pregnancy & Lactation
Femara is contraindicated during pregnancy and lactation. It can cause fetal harm. Women of childbearing potential should use effective contraception.
