Femastin-01 Vaginal Cream contains Estriol, a natural estrogen, used to treat vaginal atrophy associated with menopause. It helps restore the normal flora and physiological pH of the vagina, improving dryness, irritation, and discomfort.
Uses & Indications
Dosage
Adults
Initially, one applicator-full (approx. 0.5g cream containing 0.5mg Estriol) daily for 2-3 weeks. Then, a maintenance dose of one applicator-full twice weekly.
Elderly
No specific dosage adjustment required for elderly patients, follow adult dosage.
Renal_impairment
No specific dosage adjustment required; minimal systemic absorption.
How to Take
Administer intravaginally using the provided applicator, preferably at bedtime. Fill the applicator to the mark, insert deeply into the vagina, and depress the plunger.
Mechanism of Action
Estriol directly acts on estrogen receptors in the vaginal mucosa, leading to proliferation of vaginal epithelial cells, increased glycogen content, and restoration of the normal acidic vaginal environment. This helps improve vaginal lubrication, elasticity, and reduce symptoms of atrophy.

Side Effects
Contraindications
- •Known, suspected or past breast cancer.
- •Known or suspected estrogen-dependent malignant tumours.
- •Undiagnosed vaginal bleeding.

- •Previous idiopathic or current venous thromboembolism (DVT, PE).
- •Active or recent arterial thromboembolic disease (e.g., angina, myocardial infarction, stroke).
- •Acute liver disease or a history of liver disease where liver function tests have failed to return to normal.
- •Porphyria.
- •Hypersensitivity to the active substance or to any of the excipients.
Drug Interactions
Corticosteroids
May increase effects of corticosteroids due to potential minor systemic absorption.
No significant drug interactions are expected due to minimal systemic absorption.
However, theoretically, drugs that induce hepatic enzymes (e.g., barbiturates, carbamazepine, rifampicin) might increase estriol metabolism, but this is less relevant for topical preparations.
Storage

Overdose
Overdose is unlikely with intravaginal administration due to minimal systemic absorption. If accidental oral ingestion occurs, symptoms like nausea, vomiting, and vaginal bleeding may appear. Treatment is symptomatic and supportive.
Pregnancy & Lactation
Contraindicated during pregnancy. Not recommended during lactation as estriol is excreted in breast milk and may affect the infant.
