Description
Overview of the medicine
Clomiphene citrate is a non-steroidal fertility medicine used to stimulate ovulation in women who desire pregnancy. It works by causing the pituitary gland to produce more follicle-stimulating hormone (FSH) and luteinizing hormone (LH), which stimulate the growth and release of an egg (ovulation).
Uses & Indications
Dosage
Adults
50 mg orally once daily for 5 days, starting on day 5 of the menstrual cycle or at any time in amenorrheic patients. If ovulation does not occur, dosage may be increased to 100 mg daily for 5 days in subsequent cycles, up to a maximum of 3 cycles.
Elderly
Not typically indicated for elderly patients.
Renal_impairment
No specific dose adjustment guidelines provided, use with caution.
How to Take
Oral administration, usually starting on the 5th day of the menstrual cycle, for 5 days.
Mechanism of Action
Clomiphene is a selective estrogen receptor modulator (SERM) that binds to estrogen receptors in the hypothalamus, pituitary gland, and other estrogen-responsive tissues. By blocking these receptors, it is believed to prevent the negative feedback of estrogen, leading to an increase in GnRH, FSH, and LH secretion, which stimulates ovarian follicular growth and ovulation.
Pharmacokinetics
Onset
Ovulation typically occurs 5-10 days after the last dose of a cycle.
Excretion
Primarily excreted via feces; some via urine.
Half life
5-7 days (variable due to enterohepatic recirculation).
Absorption
Readily absorbed after oral administration.
Metabolism
Metabolized in the liver.
