Description
Overview of the medicine
Flibanserin 100 mg Tablet is a non-hormonal medication used to treat acquired, generalized hypoactive sexual desire disorder (HSDD) in premenopausal women. It helps to restore sexual desire by affecting neurotransmitters in the brain.
Uses & Indications
Dosage
Adults
100 mg orally once daily at bedtime.
Elderly
Not recommended; safety and efficacy not established in postmenopausal or elderly women.
Renal_impairment
No dose adjustment needed for mild to moderate renal impairment. Not studied in severe renal impairment.
How to Take
Take orally once daily at bedtime. Do not take with alcohol. Swallow the tablet whole with water.
Mechanism of Action
Flibanserin acts as a postsynaptic 5-HT1A receptor agonist and a 5-HT2A receptor antagonist, as well as a D4 receptor antagonist. It helps to modulate neurotransmitter activity in the brain, rebalancing the inhibitory and excitatory systems to improve sexual desire.
Pharmacokinetics
Onset
Gradual; clinical effect may take 4-8 weeks.
Excretion
Excreted primarily in feces (51%) and urine (44%) as metabolites.
Half life
Approximately 11 hours (terminal half-life).
Absorption
Rapidly absorbed; peak plasma concentrations reached within 0.5-3 hours.
Metabolism
Extensively metabolized primarily by CYP3A4 and to a lesser extent by CYP2C19.
Side Effects
Contraindications
- •Concomitant use with alcohol (risk of severe hypotension and syncope).
- •Concomitant use with moderate or strong CYP3A4 inhibitors.
- •Hepatic impairment (liver problems).
- •Known hypersensitivity to flibanserin.
Drug Interactions
Alcohol
Increases risk of severe hypotension and syncope.
Oral Contraceptives
May increase flibanserin exposure, requiring caution.
Central Nervous System (CNS) Depressants
Increased risk of sedation and dizziness.
CYP3A4 Inducers (e.g., rifampin, St. John's Wort)
Decreases flibanserin concentrations, potentially reducing efficacy.
CYP3A4 Inhibitors (e.g., ketoconazole, fluconazole, grapefruit juice)
Significantly increases flibanserin concentrations, leading to hypotension and syncope.
Storage
Store at room temperature (20°C to 25°C), away from moisture and light. Keep out of reach of children.
Overdose
Symptoms of overdose include somnolence, sedation, nausea, dizziness. Management is symptomatic and supportive, including maintaining an open airway, monitoring vital signs, and gastric decontamination if appropriate.
Pregnancy & Lactation
Not recommended during pregnancy due to potential for fetal harm (animal studies showed adverse effects). Avoid during lactation; it is unknown if flibanserin is excreted in human milk.
