Description
Overview of the medicine
Flonor 0.4 mg Capsule contains Tamsulosin Hydrochloride, an alpha-blocker used to treat symptoms of an enlarged prostate (benign prostatic hyperplasia or BPH). It helps relax the muscles in the prostate and bladder neck, making it easier to urinate.
Uses & Indications
Dosage
Adults
0.4 mg once daily, approximately 30 minutes after the same meal each day. May be increased to 0.8 mg once daily if needed, after several weeks.
Elderly
No dosage adjustment required.
Renal_impairment
No dosage adjustment required for mild to moderate renal impairment. Use with caution in severe renal impairment (CrCl < 10 mL/min).
How to Take
Take orally once daily, approximately 30 minutes after the same meal each day. Do not crush, chew, or open the capsule.
Mechanism of Action
Tamsulosin selectively blocks alpha-1A and alpha-1B adrenergic receptors in the prostate, prostatic capsule, bladder neck, and prostatic urethra. Blocking these receptors leads to relaxation of smooth muscle in these tissues, resulting in improved urine flow and reduced symptoms of BPH.
Pharmacokinetics
Onset
Clinical improvement usually seen within 1 week, but maximum benefit may take 2-4 weeks.
Excretion
Primarily excreted in the urine (about 76% as metabolites, 9% as unchanged drug), and a small amount in feces.
Half life
Approximately 9-13 hours (in plasma), 14-15 hours (terminal half-life).
Absorption
Well absorbed from the gastrointestinal tract, nearly 100% bioavailability. Absorption is delayed and slightly reduced by food.
Metabolism
Extensively metabolized by cytochrome P450 enzymes in the liver, primarily CYP3A4 and CYP2D6.
Side Effects
Contraindications
- •Hypersensitivity to Tamsulosin or any component of the formulation
- •History of orthostatic hypotension
Drug Interactions
Other alpha-adrenergic blockers
Increased risk of hypotension; avoid concomitant use.
CYP2D6 inhibitors (e.g., paroxetine)
May increase Tamsulosin exposure; use with caution.
CYP3A4 inhibitors (e.g., ketoconazole)
May increase Tamsulosin exposure; use with caution, consider dose reduction.
PDE5 inhibitors (e.g., sildenafil, tadalafil)
Increased risk of hypotension; use with caution, start with lower doses of PDE5 inhibitors.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
