Description
Overview of the medicine
Genclovir 0.15% Eye Gel contains Ganciclovir, an antiviral agent used to treat acute herpetic keratitis (dendritic ulcers) of the eye.
Uses & Indications
Dosage
Adults
One drop in the affected eye 5 times daily until re-epithelialization, then 3 times daily for 7 days.
Elderly
No specific dosage adjustment required for elderly patients.
Renal_impairment
Not applicable for ophthalmic use due to minimal systemic absorption.
How to Take
For ophthalmic use only. Tilt head back, pull down lower eyelid, and apply one drop into the conjunctival sac. Avoid touching the dropper tip to the eye or any other surface to prevent contamination. Wash hands before and after use.
Mechanism of Action
Ganciclovir is a synthetic guanine derivative that is phosphorylated to ganciclovir triphosphate by viral and cellular kinases. Ganciclovir triphosphate inhibits viral DNA synthesis by competitive inhibition of viral DNA polymerase and by direct incorporation into viral DNA, leading to chain termination.
Pharmacokinetics
Onset
Clinical improvement typically seen within 2-7 days.
Excretion
Not significantly excreted systemically after ocular application; primarily local action.
Half life
Not relevant for ophthalmic formulation due to minimal systemic absorption.
Absorption
Minimal systemic absorption after ocular administration. Local absorption into cornea and anterior chamber.
Metabolism
No significant ocular or systemic metabolism reported for ophthalmic use.
Side Effects
Contraindications
- •Hypersensitivity to ganciclovir or any component of the gel
Drug Interactions
Other ophthalmic preparations
If using other eye drops/gels, administer them at least 5 minutes apart.
Storage
Store at room temperature (15-25°C). Do not freeze. Keep out of reach of children.
Overdose
Ocular overdose is unlikely to cause systemic toxicity due to minimal absorption. If excessive amounts are applied, flush the eye with lukewarm water.
Pregnancy & Lactation
Use during pregnancy only if the potential benefit justifies the potential risk to the fetus. It is unknown if ganciclovir is excreted in human milk after ophthalmic administration; caution should be exercised when administered to a nursing mother.
