Description
Overview of the medicine
Gliclazide is an oral antidiabetic drug used to control high blood sugar levels in people with type 2 diabetes mellitus. It belongs to the sulfonylurea class of drugs.
Uses & Indications
Dosage
Adults
Initially 40-80 mg once daily, usually before breakfast. Dose can be adjusted up to 320 mg/day in divided doses.
Elderly
Similar to adult dosage, but caution with initial dosing and titration to avoid hypoglycemia.
Renal_impairment
No significant dose adjustment for mild to moderate impairment; use with caution in severe renal impairment.
How to Take
Take orally, usually once daily with breakfast. Swallow whole with water, do not crush or chew.
Mechanism of Action
Stimulates insulin secretion from pancreatic beta cells by binding to sulfonylurea receptors.

Excretion
Primarily excreted in urine (60-70%) as metabolites, with a small amount in feces.

Side Effects
Contraindications
- Type 1 Diabetes Mellitus
- Diabetic ketoacidosis
- Severe renal or hepatic impairment
- Hypersensitivity to gliclazide or other sulfonylureas
- Concomitant miconazole therapy
Drug Interactions
Miconazole
Potentiates hypoglycemic effect, leading to severe hypoglycemia.
Beta-blockers
Can mask symptoms of hypoglycemia.
Phenylbutazone, Alcohol
Increases hypoglycemic effect.
Corticosteroids, Diuretics
May decrease hypoglycemic effect.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Can cause severe hypoglycemia. Management includes oral glucose for conscious patients or intravenous glucose/glucagon for unconscious patients.
Pregnancy & Lactation
Not recommended during pregnancy or breastfeeding. Insulin is preferred for glycemic control.
