Overview of the medicine
Gyned vaginal suppository is a combination medication used for the local treatment of mixed vaginal infections caused by fungi and bacteria. It contains Clotrimazole, an antifungal agent, and Clindamycin, an antibacterial agent.
Uses & Indications
Dosage
Adults
Insert one suppository intravaginally once daily, preferably at bedtime, for 3 to 7 consecutive days depending on the severity of the infection and clinical response. Typically a 7-day course is recommended for optimal results.
Elderly
No specific dosage adjustment required for elderly patients.

No specific dosage adjustment required for renal impairment as systemic absorption is minimal.
How to Take
For intravaginal use only. Insert the suppository deep into the vagina, preferably while lying down, at bedtime. Use the applicator provided if applicable.
Mechanism of Action
Clotrimazole acts by inhibiting the biosynthesis of ergosterol, an essential component of the fungal cell membrane, leading to increased membrane permeability and leakage of cellular contents. Clindamycin inhibits bacterial protein synthesis by binding to the 50S ribosomal subunit, thus preventing peptide bond formation.

Mainly excreted via feces (Clotrimazole) and urine/feces (Clindamycin) after systemic absorption, though vaginal absorption is minimal.

Minimal systemic absorption after vaginal administration. Clotrimazole is poorly absorbed from the vagina. Clindamycin also has low systemic absorption from the vagina.

Contraindications
- Hypersensitivity to Clotrimazole, Clindamycin, or any component of the suppository
- History of regional enteritis, ulcerative colitis, or antibiotic-associated colitis
Drug Interactions
Warfarin
Clindamycin may potentiate the effects of vitamin K antagonists like warfarin, increasing bleeding risk.
Other vaginal products
Concurrent use of other intravaginal products during treatment should be avoided.
Neuromuscular blocking agents
Clindamycin has neuromuscular blocking properties and should be used with caution in patients receiving such agents.
