Description
Overview of the medicine
Gynomix Vaginal Suppository is a combination medicine containing an antifungal (Clotrimazole) and an antibiotic (Clindamycin). It is used to treat mixed vaginal infections caused by both fungi and bacteria, such as candidiasis and bacterial vaginosis.
Uses & Indications
Dosage
Adults
Insert one suppository intravaginally, preferably at bedtime, for 3-7 consecutive nights as directed by a physician.
Elderly
No specific dose adjustment required for elderly patients.
Renal_impairment
No dose adjustment necessary due to minimal systemic absorption.
How to Take
For intravaginal use only. Insert deeply into the vagina, preferably when lying down, usually at bedtime. Wash hands thoroughly before and after insertion.
Mechanism of Action
Clotrimazole inhibits ergosterol synthesis, an essential component of fungal cell membranes, leading to increased membrane permeability and leakage of cellular contents. Clindamycin binds to the 50S ribosomal subunit of susceptible bacteria, inhibiting protein synthesis and thus bacterial growth.
Pharmacokinetics
Onset
Symptom relief typically begins within a few days.
Excretion
Primarily excreted locally or via minor systemic elimination of absorbed amounts.
Half life
Not clinically significant for systemic half-life due to local action.
Absorption
Minimal systemic absorption after vaginal administration; localized action.
Metabolism
Minimally metabolized locally, not extensively studied for systemic metabolism due to low absorption.
Side Effects
Contraindications
- Hypersensitivity to Clotrimazole, Clindamycin, or any excipients
- History of regional enteritis, ulcerative colitis, or antibiotic-associated colitis
Drug Interactions
Warfarin
Although systemic absorption is minimal, clindamycin can potentiate the effects of oral anticoagulants (e.g., warfarin), leading to increased bleeding risk. Monitor INR carefully.
Spermicides
May reduce the effectiveness of certain spermicides.
Other vaginal products
Avoid concomitant use with other intravaginal products.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Due to minimal systemic absorption, overdose with vaginal suppositories is unlikely to cause systemic toxicity. Local irritation or discomfort may occur. Treatment is symptomatic and supportive.
Pregnancy & Lactation
Consult a doctor before use during pregnancy and lactation. Clotrimazole is generally considered safe in the second and third trimesters. Clindamycin vaginal preparations may be used if clearly needed and benefits outweigh risks. Minimal systemic absorption reduces risks.
