Imatinib is a targeted therapy used to treat certain types of cancer, primarily chronic myeloid leukemia (CML) and gastrointestinal stromal tumors (GIST). It works by blocking specific proteins that promote cancer cell growth.
Uses & Indications
Dosage
Adults
For CML (chronic phase): 400 mg once daily. For GIST: 400 mg once daily. Dosage may be adjusted based on response and tolerability, up to 800 mg daily in divided doses for some indications.
Elderly
No specific dose adjustment generally required, but monitor for adverse effects.
Renal_impairment
Patients with severe renal impairment should start at a reduced dose (e.g., 300 mg daily for CML chronic phase) and be monitored closely.
How to Take
Take orally with a meal and a large glass of water to minimize gastrointestinal irritation. Do not crush or cut tablets.
Mechanism of Action
Imatinib selectively inhibits the Bcr-Abl tyrosine kinase, which is characteristic of chronic myeloid leukemia and acute lymphoblastic leukemia. It also inhibits the receptor tyrosine kinases for platelet-derived growth factor (PDGF) and stem cell factor (SCF), c-KIT. These inhibitions lead to the suppression of proliferation and induction of apoptosis in various cancer cell lines.
