Description
Overview of the medicine
Inolac 335 gm oral solution contains Macrogol 3350 (Polyethylene Glycol), an osmotic laxative used for the treatment of chronic constipation and occasionally for bowel preparation before medical procedures. It works by increasing the water content in the stool, making it softer and easier to pass.
Uses & Indications
Dosage
Adults
For chronic constipation: 1 sachet (335 gm) dissolved in 125 mL of water, taken once daily. Dose may be adjusted according to response, from 1 sachet every other day to 2 sachets daily. For bowel preparation: Specific regimen as directed by physician.
Elderly
No specific dose adjustment is generally required, but caution is advised in frail elderly patients.
Renal_impairment
No dose adjustment is generally required as systemic absorption is negligible.
How to Take
Dissolve the contents of one sachet in 125 mL (approximately half a glass) of water. Stir well until all the powder has dissolved, then drink. The solution can be taken with or without food.
Mechanism of Action
Macrogol 3350 is an inert, long-chain polymer that acts as an osmotic agent. It passes through the gastrointestinal tract largely unabsorbed and un-metabolised. It retains water in the bowel lumen, increasing the stool volume and softening it, which stimulates colonic peristalsis and facilitates bowel evacuation.
Pharmacokinetics
Onset
Typically starts to work within 24-48 hours.
Excretion
Excreted largely unchanged in the faeces.
Half life
Not applicable due to minimal systemic absorption.
Absorption
Minimally absorbed from the gastrointestinal tract (less than 1%).
Metabolism
Not metabolised by gut bacteria or human enzymes.
Side Effects
Contraindications
- Intestinal obstruction or suspicion of intestinal obstruction
- Perforation of the gastrointestinal tract
- Severe inflammatory bowel disease (e.g., Crohn's disease, ulcerative colitis)
- Toxic megacolon
Drug Interactions
Other oral medications
May reduce the absorption of other orally administered medications if taken simultaneously. It is advisable to take other medications at least one hour before or after Macrogol 3350.
Storage
Store below 25°C, in a dry place. Keep out of reach of children. Do not use after the expiry date.
Overdose
Overdose may lead to severe diarrhea and electrolyte disturbances. Treatment involves symptomatic and supportive care, including electrolyte monitoring and replacement if necessary.
Pregnancy & Lactation
Generally considered safe for use during pregnancy and lactation as systemic absorption is minimal. However, consult a healthcare professional before use.
