Description
Overview of the medicine
Ketoalfa 600 mg Tablet contains Ketoprofen, a non-steroidal anti-inflammatory drug (NSAID) used to relieve pain, inflammation, and fever. It is commonly prescribed for conditions like arthritis, muscle pain, and menstrual cramps.
Uses & Indications
Dosage
Adults
For sustained-release 600 mg tablets: 600 mg once daily. For immediate-release: 50-100 mg 2-3 times daily, maximum 300 mg/day (consult specific product labeling as 600mg is often a sustained release dose).
Elderly
Lower doses should be considered; closely monitor renal function and potential for adverse effects.
Renal_impairment
Significant dose reduction may be required. Avoid in severe renal impairment (creatinine clearance <30 mL/min).
How to Take
Take orally with food or milk to minimize gastrointestinal upset. Do not crush or chew sustained-release tablets.
Mechanism of Action
Ketoprofen works by inhibiting the activity of cyclooxygenase (COX) enzymes, COX-1 and COX-2. This inhibition reduces the production of prostaglandins, which are chemicals responsible for pain, inflammation, and fever.
Pharmacokinetics
Onset
Pain relief typically begins within 30-60 minutes.
Excretion
Mainly through the urine (60-90% as metabolites), with a small amount excreted in feces.
Half life
Approximately 1-4 hours (for immediate release formulations).
Absorption
Rapidly and well absorbed from the gastrointestinal tract; peak plasma concentrations reached within 0.5-2 hours.
Metabolism
Primarily hepatic, largely by conjugation with glucuronic acid.
Side Effects
Contraindications
- •Hypersensitivity to Ketoprofen, aspirin, or other NSAIDs
- •Active gastrointestinal bleeding or peptic ulcer disease
- •Severe renal or hepatic impairment
- •Severe heart failure
- •Third trimester of pregnancy
Drug Interactions
Lithium
Increased plasma lithium levels, leading to toxicity.
Methotrexate
Increased plasma methotrexate levels, leading to toxicity.
Corticosteroids
Increased risk of gastrointestinal ulceration and bleeding.
Diuretics and ACE inhibitors
Reduced efficacy of these drugs and increased risk of renal impairment.
Warfarin and other anticoagulants
Increased risk of bleeding.
Storage
Store below 30°C in a dry place, protected from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose may include nausea, vomiting, epigastric pain, lethargy, drowsiness, and gastrointestinal bleeding. Management is symptomatic and supportive; there is no specific antidote.
Pregnancy & Lactation
Avoid in the third trimester of pregnancy due to risk of premature closure of the ductus arteriosus and renal dysfunction in the fetus. Use during early pregnancy and lactation only if clearly needed and benefits outweigh risks, under medical supervision.
