Description
Overview of the medicine
Lansoprazole is a proton pump inhibitor used to treat stomach and esophageal problems like acid reflux (GERD) and ulcers. It works by decreasing the amount of acid your stomach makes.
Uses & Indications
Dosage
Adults
Duodenal ulcer: 30 mg once daily for 4 weeks. Gastric ulcer: 30 mg once daily for 8 weeks. Reflux esophagitis: 30 mg once daily for 4-8 weeks.
Elderly
No dosage adjustment generally required, but caution is advised in patients with severe hepatic impairment.
Renal_impairment
No dosage adjustment required.
How to Take
Take capsule whole, do not crush or chew. Should be taken before meals, preferably in the morning.
Mechanism of Action
Lansoprazole is a proton pump inhibitor that suppresses gastric acid secretion by specific inhibition of the H+/K+-ATPase enzyme system at the secretory surface of the gastric parietal cell.
Pharmacokinetics
Onset
Within 1-2 hours.
Excretion
Primarily excreted in urine (approximately 33%) and feces (approximately 67%).
Half life
1.3 to 1.7 hours.
Absorption
Rapidly absorbed after oral administration. Bioavailability is about 80-90%.
Metabolism
Extensively metabolized in the liver, primarily by CYP2C19 and CYP3A4.
Side Effects
Contraindications
- Hypersensitivity to lansoprazole or any component of the formulation
- Concomitant use with atazanavir
Drug Interactions
Warfarin
Increased INR/bleeding risk
Sucralfate/Antacids
Reduced bioavailability of lansoprazole; take lansoprazole 30 minutes before antacids/sucralfate
Ketoconazole, Itraconazole
Decreased absorption due to altered gastric pH
Storage
Store in a cool, dry place, away from light. Do not store above 30°C.
Overdose
No specific antidote. Symptomatic and supportive treatment. Lansoprazole is not significantly removed by hemodialysis.
Pregnancy & Lactation
Pregnancy Category B. Use during pregnancy only if clearly needed. Exercise caution in breastfeeding as it is excreted in breast milk.
